目的:研究忍冬藤颗粒治疗急性痛风性关节炎模型大鼠抗炎消肿止痛的效果并对其机理进行探讨。方法:给予大鼠乙胺丁醇(189mg/kg)和腺嘌呤(1.26g/kg)灌胃,每天1次连续3周诱导高尿酸血症模型,然后在此基础上以大鼠的右踝关节外侧后方为穿刺点,针尖斜向前上方与胫骨成45°角刺入关节腔,每只注射0.2ml尿酸钠溶液,连续3天。同时各干预组大鼠分别灌胃忍冬藤颗粒23.8g/kg、11.9g/kg、5.95g/kg,别嘌呤醇0.455g/kg(第3周开始,连续1周),秋水仙碱18.9mg/kg(第4周开始.连续1周)进行干预。实验结束后,观察忍冬藤颗粒对模型大鼠关节肿胀、步态、炎症的影响,检测大鼠关节软骨中的IL-1β的表达水平。结果:与空白组比较,模型组大鼠关节肿胀增加(6.20±0.19),关节炎症(5.25±1.04)、跛行步态评分(5.25±1.04)增高,关节软骨IL-1β表达水平上升(12.33±1.36);与模型组比较,药物干预24h、48h、72h后,忍冬藤颗粒各组大鼠关节肿胀度周径明显减小,其中忍冬藤颗粒11.9g/kg剂量组对关节肿胀度的疗效更好(4.51±0.25);与模型组比较,给药后72h后,秋水仙碱(18.9mg/kg)+别嘌呤醇(0.455g/kg)组可以明显降低关节炎症程度,给药48、72h后,忍冬藤各剂量组可以明显降低关节炎症评分,其中忍冬藤颗粒11.9g/kg剂量组降低炎症评分效果更优(1.00±1.07);与模型组比较,给药后24h、48h、72h后,忍冬藤颗粒各剂量组可以明显降低步态评分(1.00±1.07);与模型组比较,秋水仙碱(18.9mg/kg)+别嘌呤醇(0.455g/kg)、忍冬藤颗粒各剂量组大鼠血清IL-1β表达明显降低,其中忍冬藤颗粒11.9g/kg(6.16±0.70)、23.8g/kg(6.96±0.79)剂量组IL-1β表达降低效果最优。结论:忍冬藤颗粒可以起到抗炎消肿止痛的功效,对急性痛风性关节炎模型大鼠有着明显的治疗作用,其作用机制可能是抑制炎症关节IL-1β致炎因子的生成而实现的。

Compared with the model group, the levels of inflammatory markers decreased significantly after treatment with 11.9g / kg Houttuynia Granules group at 24h, 48h, 72h , The honeysuckle particles in each dose group can significantly reduce the gait score (1.00 ± 1.07); compared with the model group, colchicine (18.9mg / kg) + allopurinol (0.455g / kg) The expression of IL-1β in the serum of rats was significantly decreased, and the expression of IL-1β in the group of honeysuckle particles of 11.9g / kg (6.16 ± 0.70) and 23.8g / kg (6.96 ± 0.79) Best effect. Conclusion: Honeysuckle Granules can play an anti-inflammatory, detumescent and analgesic effect. It has a significant therapeutic effect on rats with acute gouty arthritis and its mechanism may be to inhibit the production of IL-1β-induced inflammatory cytokines .
