目的:运用高效液相色谱-高分辨飞行时间质谱(HPLC-TOF/MS)联用技术快速鉴别肝力保胶囊中化学成分。方法:色谱分离采用SHISEIDO MG C18色谱柱(3.0 mm×100 mm,3μm);流动相0.1%甲酸水(A)-乙腈(B),梯度洗脱(0~10min,5%~20%B;10~30 min,20%~70%B),柱温25℃,流速0.6 m L·min-1,柱后分流比2∶1。质谱定性采用飞行时间质谱,电喷雾离子源(ESI),正负离子模式扫描(m/z 100~1 000)。结果:一次性鉴别出肝力保胶囊中30种化学成分。结论:该研究比较全面地阐明了肝力保胶囊的化学组成,为该复方的药效学物质基础研究和质量控制奠定了基础。
OBJECTIVE: To rapidly identify the chemical constituents in Heleng capsule by using high performance liquid chromatography-high resolution liquid chromatography-time of flight mass spectrometry (HPLC-TOF / MS). METHODS: The chromatographic separation was carried out on a SHISEIDO MG C18 column (3.0 mm × 100 mm, 3 μm). The mobile phase consisted of 0.1% formic acid in water and acetonitrile, gradient elution (0-10 min, 5% -20% 10 ~ 30 min, 20% ~ 70% B). The column temperature was 25 ℃ and the flow rate was 0.6 m L · min-1. The post-column split ratio was 2:1. MS was qualitatively characterized by time of flight mass spectrometry, electrospray ionization (ESI), positive and negative ion mode scanning (m / z 100-1000). Results: A one-time identification of liver capsule 30 kinds of chemical composition. CONCLUSION: This study comprehensively elucidates the chemical composition of Ganoderma capsule and paves the way for the basic research and quality control of this compound.
