利用实时荧光定量PCR检测经过ASA处理的萝芙木毛状根与对照组中阿玛碱合成途径相关基因的相对表达量,同时使用HPLC分别测定阿玛碱质量分数,并对其相互关系进行分析,研究发现:阿玛碱合成下游途径基因(STR,SGD)相对表达量的变化与阿玛碱质量分数变化一致,表明阿玛碱生物合成受到这些基因特别是STR的严谨调控;MEP途径受到ASA作用,相关基因表达量提高,但阿玛碱质量分数反而降低,有可能是因为ASA导致了阿玛碱的细胞外分泌所致.
Real-time fluorescent quantitative PCR was used to detect the relative expression of the genes related to the pathogen of armaine in the hairy roots of Rauwolfia treated with ASA and the control group, and the correlation coefficients were determined by HPLC The results showed that the changes of the relative expression levels of arginine synthase downstream pathway genes (STR, SGD) were in accordance with the changes of the concentration of armaine, indicating that arbusid biosynthesis was strictly regulated by these genes, especially STR. The MEP pathway was regulated by ASA Effect, related gene expression increased, but the lower the quality of the ammamine, may be due to the ASA led to the extracellular secretion of armase.
