目的 :通过对犬下颌骨牵张成骨过程中咀嚼肌肌球蛋白重链亚型mRAN含量变化的测定 ,研究牵张成骨过程中肌肉改建的过程及能力。方法 :12只西安本地杂种犬分为 6组 :对照组、牵张 5d组、牵张 10d组、固定 2周组、固定 4周组和固定 8周组。麻醉后分别取嚼肌与颏舌骨肌的中央部分对MHCⅠ、MHCⅡ及GAPDH(内参照 )的mRNA进行RT -PCR。结果 :试验各组嚼肌中各项检测指标与对照组相比没有显著性差异。牵张 5d后颏舌骨肌中MHCⅠ的mRNA相对含量开始上升 ,MHCⅡ的mRNA相对含量开始下降 ,MHCⅠ /MHCⅡ之比增加 ,刚刚牵张完成时变化最为明显 ,牵张完成固定 4~ 8周时基本恢复。结论 :在犬下颌骨牵张成骨过程中相关的咀嚼机会发生组份的改变 ,并且这种改建有利于牵张成骨术的术后稳定
OBJECTIVE: To study the process and ability of muscle remodeling during distraction osteogenesis by measuring the change of mRAN content of the myosin heavy chain subtype in the process of mandibular distraction osteogenesis in dogs. Methods: Twelve Xian local hybrid dogs were divided into 6 groups: control group, stretch 5d group, stretch 10d group, fixed 2 week group, fixed 4 week group and fixed 8 week group. After anesthesia, the medial portions of the muscles of the chewing muscles and genioglossus were respectively subjected to RT-PCR for mRNAs of MHC I, MHC II and GAPDH (internal reference). Results: There was no significant difference between the test items in each group and the control group. The relative content of MHC Ⅰ mRNA in genioglossus muscle began to increase after 5 days of distraction, the relative content of MHC Ⅱ mRNA began to decrease, and the ratio of MHCⅠ / MHCⅡ increased. At the time of distraction, the change was most obvious, and the stretch was fixed for 4 to 8 weeks Basic recovery. CONCLUSIONS: Changes in the composition of components associated with chewing during the process of distraction osteogenesis of canine mandibles are associated with postoperative stabilization of distraction osteogenesis
