目的 研究人CD2 2 6 (PTA1)启动子及其 5’上游调控序列。方法 应用计算机软件预测启动子序列 ,并分析人CD2 2 6基因 5’上游调控序列以及通过RT -PCR的方法研究开放读框中的单核苷酸多态性。结果 CD2 2 6基因在 -375bp处和 - 130bp处可能有两个启动子 ,含有AP - 1、Ets- 1、Sp1、P30 0、PU .1、PEA3等转录因子结合序列 ,在编码胞浆区的序列中存在一个单核苷酸多态性位点。结论 CD2 2 6基因表达受到多种转录因子和 5’端上游调控序列的调控。
Objective To study the human CD2 2 6 (PTA1) promoter and its 5 ’upstream regulatory sequence. Methods The promoter sequence was predicted by computer software. The 5 ’upstream regulatory sequence of human CD2 2 6 gene was analyzed and the single nucleotide polymorphism in the open reading frame was studied by RT-PCR. Results There were two promoters at -375bp and -130bp in CD2 2 6 gene, including transcription factor binding sequences of AP - 1, Ets1, Sp1, P30 0, PU1 and PEA3. There is a single nucleotide polymorphism site in the sequence. Conclusion CD2 2 6 gene expression is regulated by many transcription factors and 5 ’upstream regulatory sequences.
