目的:天然抗氧化剂的抗肿瘤作用已逐步被认识。用茶多酚、丹酚酸B和知母宁三种天然抗氧化剂进行MTT实验,观察和比较它们对HL-60细胞生长和增殖的影响。试图为系统研究天然抗氧化剂的抗癌作用、自由基与肿瘤的关系提供一些实验依据。方法:取对数生长的HL-60细胞,以1×108/L细胞浓度接种于5mL培养瓶中,同时加药物培养。3种药物均设5个剂量组(终浓度5~80mg/L)及空白对照组(乙醇)和阳性对照组(维甲酸)。药物处理4d后取细胞进行MTT实验(5个平行孔),计算细胞相对存活率。实验结果经方差分析、各实验组与对照组均值比较,并计算IC50及95%可信区间。结果:实验结果显示,在体外,茶多酚、丹酚酸B和知母宁在5~80mg/L浓度范围内对HL-60细胞存活和增殖均有一定的抑制作用,且呈明显的剂量依赖关系。茶多酚、丹酚酸B和知母宁的IC50及95%可信区间分别为17.7mg/L、12.7~24.8mg/L,22.5mg/L、15.5~32.6mg/L和17.1mg/L、11.9~24.5mg/L。结论:茶多酚、丹酚酸B和知母宁三种天然抗氧化剂对HL-60细胞生长的抑制作用,可能与其抗氧化性质有关。虽然,它们的IC?
Objective: The anti-tumor effects of natural antioxidants have been gradually recognized. MTT assays were performed with three polyphenols, salvianolic acid B and Zhimulin, and their effects on the growth and proliferation of HL-60 cells were observed and compared. Attempts to systematically study the anticancer effects of natural antioxidants and the relationship between free radicals and tumors provide some experimental evidence. METHODS: Logarithmic growth of HL-60 cells was performed and inoculated into 5 mL flasks at a cell concentration of 1×108/L. All three drugs were given 5 dose groups (final concentration 5 to 80 mg/L), blank control group (ethanol) and positive control group (retinoic acid). After 4 days of drug treatment, cells were taken for MTT assay (5 parallel wells) and the relative cell survival rate was calculated. The experimental results were compared by analysis of variance, the average value of each experimental group and the control group, and the IC50 and 95% confidence interval were calculated. Results: The experimental results showed that in vitro, tea polyphenols, salvianolic acid B and Zhim’nin have a certain inhibitory effect on the survival and proliferation of HL-60 cells in the range of 5-80 mg/L.
