目的以FAT/CD36为切入点,研究母体孕期炎症刺激对子代脂质代谢的影响。方法 8周龄C57小鼠,♀♂2∶1合笼配种,d 2♀鼠分笼饲养记为在孕0 d,在孕11 d给予孕鼠一次性腹腔注射脂多糖(LPS)(75μg·kg~(- 1)),对照组注射0.2 m L的生理盐水。分别于子鼠4、8、12周取材(♀鼠取肾周脂肪、♂鼠取附睾周围脂肪),对子代小鼠体重、内脏脂肪重量、脂肪组织和细胞中游离脂肪酸(FFA)、甘油三酯(TG)、FAT/CD36表达量进行检测。结果与NS组相比,LPS组小鼠体重、内脏脂肪重量、脂肪系数明显增高,♂鼠附睾脂肪和血液中TG、FFA的表达量明显升高;各周龄♂鼠脂肪组织和诱导的3T3-L1细胞中FAT/CD36明显上调(P<0.05)。结论母体孕期炎症刺激导致子代小鼠脂肪发育异常,游离脂肪酸(FFA)、甘油三酯(TG)、FAT/CD36的表达量明显升高,其机制可能与FAT/CD36对脂质代谢的调节有关。
Objective To study the effect of maternal inflammatory stimuli on lipid metabolism in offspring with FAT / CD36 as an entry point. Methods C57 mice of 8 weeks old were bred with 2: 1 cohorts and d 2 mice were housed in a cage for 0 d. The mice were given intraperitoneal injection of lipopolysaccharide (LPS) (75 μg · kg ~ (-1)), the control group injected 0.2 m L of saline. The rats were sacrificed at 4, 8, and 12 weeks of age respectively, and the perirenal adipose tissue was obtained from the offspring of the mice and the weight of visceral fat, free fatty acids (FFA), glycerol Triester (TG), FAT / CD36 expression levels were detected. Results Compared with NS group, the body weight, visceral fat weight and fat coefficient in LPS group were significantly increased, while the expression of TG and FFA in epididymal fat and blood were significantly increased in the LPS group. The adipose tissue and induced 3T3 FAT / CD36 was significantly up-regulated in -L1 cells (P <0.05). Conclusion The maternal inflammatory stimuli during pregnancy lead to abnormal fat development in the offspring mice. The expression of free fatty acids (FFA), triglycerides (TG) and FAT / CD36 in mice is significantly increased, which may be related to the regulation of lipid metabolism by FAT / CD36 related.
