目的:测定流感病毒HA基因序列,从分子水平分析丽水市输入性甲型H1N1流感病毒株HA基因特点。方法:Real-time PCR检测甲型H1N1流感病毒核酸,RT-PCR测定HA基因核苷酸序列。结果:测序获得位于流感病毒HA基因436 bp~1701 bp位置、长度为1266 bp的核苷酸片段。多序列比对显示A/Lishui/01/2009(H1N1)株与北美人感染猪流感A/California/04/2009(H1N1)株序列的相似性最高(nt/aa,99.8/99.3%);与其他甲型H1N1流感病毒株序列相似性均比较高(nt/aa,99.4~99.9%/99.1~99.8%),与季节性甲H1型流感代表株A/Brisbane/59/2007(H1N1)序列相似性最低(nt/aa,76.4/80.0%)。种系分析显示A/Lishui/01/2009(H1N1)株与其他甲型H1N1流感病毒,北美猪流感病毒均位于同一进化分支,亲缘关系最近,与季节性甲H1流感代表株A/Brisbane/59/2007(H1N1)进化关系最远。结论:丽水输入性流感病毒A/Lishui/01/2009(H1N1)株属于甲型H1N1流感病毒,与北美猪流感病毒同源并隶属于传统猪流感病毒。
OBJECTIVE: To determine the HA gene sequence of influenza virus and analyze the HA gene characteristics of imported H1N1 influenza virus strain in Lishui City from the molecular level. Methods: Real-time PCR was used to detect the influenza A (H1N1) virus nucleic acid and the HA gene nucleotide sequence was determined by RT-PCR. Results: The nucleotide sequence of the HA gene of influenza virus HA located between 436 bp and 1701 bp in length and 1266 bp in length was obtained. Multiple sequence alignment showed the highest similarity (nt / aa, 99.8 / 99.3%) between the A / Lishui / 01/2009 (H1N1) strain and the North American human swine influenza A / California / 04/2009 The sequences of other influenza A (H1N1) strains were similar (nt / aa, 99.4-99.9% / 99.1-99.8%), similar to the sequence of A / Brisbane / 59/2007 (H1N1) Sex lowest (nt / aa, 76.4 / 80.0%). Phylogenetic analysis showed that the A / Lishui / 01/2009 (H1N1) strain was located in the same phylogenetic relationship with other Influenza A (H1N1) and North American swine influenza viruses and has the closest genetic relationship with the A / Brisbane / 59 / 2007 (H1N1) has the longest evolutionary relationship. CONCLUSION: The strain of Lishui-imported influenza A / Lishui / 01/2009 (H1N1) belongs to the type A H1N1 influenza virus and is homologous to the swine influenza virus of North American origin and belongs to the traditional swine flu virus.
