[目的]应用基因芯片技术研究青少年特发性脊柱侧弯(AIS)来源的骨髓间充质干细胞(BM-MSCs)成脂分化过程中差异表达基因,探究AIS的发病机制。[方法]分离培养AIS患儿及正常对照者BM-MSCs培养至第五代(P5),成脂诱导2周后利用基因芯片技术筛选差异表达基因,并利用RT-PCR技术验证芯片结果。[结果]在AIS及正常来源的BM-MSC成脂分化中共新发现229个基因存在显著差异表达。这些差异表达基因具有细胞粘附、转录调节及信号转导等多种功能。共新发现6条具有显著统计学意义的相关分子通路。[结论]AIS患儿BM-MSCs在成脂分化过程中的基因表达模式与正常对照相比存在明显差异。本研究新发现的差异表达基因及通路可能与AIS的发病相关,对其病因学研究具有重要意义,值得深入研究。
[Objective] To investigate the differential expression of differentially expressed genes in adipogenic differentiation of adolescent idiopathic scoliosis (BM) derived bone marrow mesenchymal stem cells (BM-MSCs) using gene chip technology and to explore the pathogenesis of AIS. [Methods] BM-MSCs isolated from cultured AIS and normal controls were cultured to the fifth generation (P5). Two weeks after the induction of adipogenic differentiation, differentially expressed genes were screened by gene chip technology and the chip results were verified by RT-PCR. [Results] A total of 229 new genes were found to be significantly differentially expressed in adipogenic differentiation of AIS and normal BM-MSCs. These differentially expressed genes have many functions such as cell adhesion, transcription regulation and signal transduction. A total of 6 new molecular pathways were found with significant statistical significance. [Conclusion] The gene expression pattern of BM-MSCs in AIS patients during adipogenic differentiation has obvious difference compared with the normal control. The differentially expressed genes and pathways newly discovered in this study may be related to the pathogenesis of AIS, which is of great significance for the study of their etiology and deserves further study.
