目的:观察生血宁抗红细胞衰老及对再生障碍性贫血的治疗作用。方法:体外氧化衰老和自然衰老评价生血宁的抗红细胞衰老作用,检测指标包括红细胞溶血率、Na-K-ATP酶、超氧化物歧化酶(SOD)、丙二醛(MDA)、红细胞膜渗透脆性、流动性及通透性、造血祖细胞集落生成。苯诱导的再生障碍性贫血模型观察生血宁对小鼠再生障碍性贫血的治疗作用,检测指标包括外周血红细胞(RBC)、网织红细胞(Ret)和血红蛋白(Hb)及血清红细胞生成素(EPO)、干细胞因子(SCF)、MDA和SOD。结果:生血宁能提高红细胞膜Na-K-ATP酶活性,降低MDA含量,防止红细胞的自然老化和氧化老化,显著降低红细胞膜渗透脆性及通透性,提高红细胞膜流动性。此外生血宁能刺激CFU-E、CFU-GM和CFU-F造血祖细胞集落生成。在再生障碍性贫血小鼠模型中,生血宁(400、200、100 mg/kg)呈剂量依赖性升高红细胞计数,显著提高红细胞血红蛋白含量和小鼠网织红细胞数,增加血清干细胞因子SCF、降低MDA含量。结论:生血宁有较强的抗红细胞衰老作用,对苯诱导的小鼠再生障碍性贫血有显著疗效。
Objective: To observe the anti-aging effect of Shengxuening on erythrocytes and the therapeutic effect on aplastic anemia. Methods: The anti-erythrocyte senescence effect of Shengxuening was evaluated by oxidative aging and natural aging in vitro. The detection indexes include hemolysis rate, Na-K-ATPase, superoxide dismutase (SOD), malondialdehyde (MDA) Fragility, mobility and permeability, hematopoietic progenitor colony formation. Benzene-induced aplastic anemia model was used to observe the therapeutic effect of Shengxuening on aplastic anemia in mice. The detection indexes include peripheral blood red blood cell (RBC), reticulocyte (Ret), hemoglobin (Hb) and serum erythropoietin ), Stem cell factor (SCF), MDA and SOD. Results: Shengxuening could increase the Na-K-ATPase activity of erythrocyte membrane, decrease the content of MDA, prevent the natural aging and oxidative aging of erythrocytes, reduce the infiltration fragility and permeability of erythrocyte membrane, and improve the fluidity of erythrocyte membrane. In addition, Sheng-Ning can stimulate CFU-E, CFU-GM and CFU-F hematopoietic progenitor colony formation. In the aplastic anemia mouse model, Shengxuening (400, 200, 100 mg / kg) increased the number of erythrocytes in a dose-dependent manner, significantly increased the hemoglobin content of red blood cells and reticulocyte count in mice, increased the serum levels of SCF, Reduce MDA content. Conclusion: Shengxuening has strong anti-erythrocyte senescence effect, benzene-induced aplastic anemia in mice has a significant effect.
