目的对冬凌草内生真菌进行筛选鉴定,并研究其体外抗肿瘤活性,以期得到冬凌草甲素高产抗肿瘤活性菌株。方法采用TLC法和HPLC法对前期从冬凌草中分离出的256株内生真菌进行筛选,HPLC-MS测定菌株发酵液中的冬凌草甲素。结合形态学特征、ITS序列对筛选出的菌株进行鉴定。MTT法测定筛选菌株的体外抗肿瘤活性。结果从冬凌草中分离出的256株内生真菌经筛选,获得1株冬凌草甲素高产菌株M-J-5,鉴定为青霉属草酸青霉菌Penicillium oxalicum Currie et Thom,其对人乳腺癌细胞系MCF-7细胞活性具有一定抑制作用。结论从冬凌草内生真菌中筛选得到产冬凌草甲素且具抗肿瘤活性的菌株M-J-5。
Objective To screen and identify Endophytic Fungus of Rubescens indica L. and to study its antitumor activity in vitro in order to obtain a highly active antitumor activity strain of Rubescensine A. Methods Twenty-six strains of endophytic fungi were isolated from Rubescens indica L. by TLC and HPLC methods. Oridonin in the fermentation broth of the strains was determined by HPLC-MS. According to the morphological characteristics, the ITS sequences were used to identify the selected strains. In vitro anti-tumor activity of the selected strains by MTT assay. Results A total of 256 endophytic fungi isolated from Rubescens indica L. were screened to obtain one high-yielding oridrin-producing strain MJ-5, which was identified as Penicillium oxalicum Currie et Thom, Cell line MCF-7 cell activity has a certain inhibitory effect. Conclusion The strain M-J-5, which produces oridonin and has antitumor activity, was screened from the endophytic fungi of Rubescens.
