目的 观察苯甲酸雌二醇对6 0 Coγ射线引起的小鼠胸腺、脾脏细胞凋亡率的影响及脾脏T、B淋巴细胞表面分子表达的改变 ,探讨其对小鼠免疫细胞凋亡的调节作用及其作用机理。方法 采用6 0 Coγ射线 4Gy单次全身照射 ,用DNA琼脂糖凝胶电泳、PI染色及双荧光流式细胞术研究小鼠免疫细胞凋亡 ,细胞表面CD95、CD2 5、CD69的表达变化规律。结果 注射照射组各细胞亚群凋亡率较单纯照射组明显降低 ,照射后 4h未出现照射组所呈现的CD95高表达。射线使脾脏细胞CD2 5和CD69表达升高 ,而注射照射组未观察到CD2 5和CD69的高表达。结论 苯甲酸雌二醇可阻断免疫细胞的激活进而阻断Fas(CD95 )系统介导的细胞凋亡途径。
Objective To observe the effect of estradiol benzoate on the apoptosis rate of thymus and spleen cells induced by 60 Co γ-rays in mice and the molecular expression of T and B lymphocytes in spleen, and to explore its regulation on the apoptosis of mouse immune cells And its mechanism of action. Methods Immunohistochemistry was used to determine the expression of CD95, CD25 and CD69 on the surface of mouse immune cells by DNA agarose gel electrophoresis, PI staining and double fluorescence flow cytometry. Results The apoptotic rates of various cell subsets in the irradiated group were significantly lower than those in the pure irradiated group, and no expression of CD95 was observed in the irradiated group 4h after irradiation. Rays increased the expression of CD25 and CD69 in spleen cells, whereas no high expression of CD25 and CD69 was observed in the irradiated group. Conclusion Estradiol benzoate can block the activation of immune cells and block the apoptotic pathway mediated by Fas (CD95) system.
