目的在于观察体外培养条件下,库普弗细胞(KC)贮脂细胞(FSC)对肝细胞(PC)基质生成调节效应及PC增殖及合成功能的影响。采用FSC、KC和PC的分离培养,观察KC和FSC对PC增殖活性和DNA含量的影响,应用免疫细胞化学和图像分析系统对PC中Ⅰ、Ⅱ、Ⅳ型胶原和纤维粘连蛋白(FN)进行定量分析,以及用液闪法检测PC中3H-脯氨酸掺入量。结果证明FSCncm可明显促进PCDNA合成,PC可合成Ⅰ、Ⅲ、Ⅳ型胶原及FN。FSCncm可促使双核的PC增多,线粒体分裂。提示肝细胞可合成Ⅰ、Ⅲ、Ⅳ型胶原及FN;PC内可能直接组装胶原纤维;KCncm、KCtcm、FSCtcm均可上调PC的基质合成能力。
The purpose of this study was to investigate the effects of Kupffer cells (KCs) -stimulating cells (FSC) on the regulation of hepatocyte (PC) stroma generation and PC proliferation and synthesis in vitro. The effects of KC and FSC on PC proliferation activity and DNA content were observed by the isolation and culture of FSC, KC and PC. Immunocytochemistry and image analysis system were used to detect the expression of collagen type Ⅰ, Ⅱ, Ⅳ and fibronectin (FN) in PC Quantitative analysis and 3H-proline incorporation in PC were determined by liquid-flash method. The results show that FSCncm can significantly promote the synthesis of PCDNA, PC can synthesize type Ⅰ, Ⅲ, Ⅳ collagen and FN. FSCncm can promote binucleate PC increased mitochondrial division. These results suggest that type Ⅰ, type Ⅲ and type Ⅳ collagen and FN can be synthesized in hepatocytes. Collagen fibers may be directly assembled in PC. KCncm, KCtcm and FSCtcm may upregulate matrix synthesis of PC.
