目的:观察痛风合剂及其优化组方对尿酸盐所致急性痛风性关节炎大鼠的疗效,明确痛风合剂优化方的疗效是否更确切。方法:选取wista大鼠54只,随机分为9组。正常对照组、模型组、西药组、痛风合剂高、中、低浓度组、痛风合剂优化方高、中、低浓度组,分别用生理盐水、吲哚美辛混悬剂、痛风合剂高、中、低浓度汤剂、痛风合剂优化方高、中、低浓度汤剂灌胃7天,早晚各1次。于灌胃第四天把25g/mL(0.05mL)浓度的尿酸盐溶液注射到大鼠踝关节腔内,制备痛风性关节炎模型。对照组大鼠关节腔注射0.05mL生理盐水。分别于灌胃前和造模前、造模后24、48、72h秤取大鼠体重;造模前一天上午和下午,造模后0、2、4、6、8、10、12、24、48、72h测取右侧距小腿关节同一部位周径(取均值);造模后72h取血,观察血浆白细胞计数,检测血清白细胞介素-1β(Inter-leukin-1β,IL-1β)的含量;取血后将大鼠处死,取踝关节,10%福尔马林固定待做病理。结果:54只大鼠均进入结果分析。灌胃前体重无差异,造模后第1、2天痛风合剂及优化方高浓度组、西药组高于模型组,存在差异;造模前一天大鼠上午和下午踝关节周径无差异,造模后大鼠关节肿胀度各组均有所增高,其中以模型组最为明显,药物治疗后,用药组肿胀度均下降低,高浓度组和吲哚美辛效果相当;痛风合剂及优化方各剂量组血浆白细胞计数和血清白细胞介素1β水平明显高于对照组且以模型组最为显著,与模型组相比,痛风合剂及优化方高浓度组差异明显;病理组织学结果显示,痛风合剂及优化方可减轻痛风性关节大鼠踝关节的组织水肿和炎性细胞浸润,改善滑膜增生,且浓度越高作用越明显。但以上检测指标中,痛风合剂组和优化方组无差异。结论:痛风合剂及优化方可以有效抑制急性痛风性关节炎发作,且该作用呈一定的剂量依赖性,但该作用在痛风合剂和优化方之间并无差异。
Objective: To observe the effect of Gout Mixture and its optimized prescription on urate-induced acute gouty arthritis in rats, and to determine whether the therapeutic effect of Gout Mixture Prescription is more accurate. Methods: 54 wistar rats were randomly divided into 9 groups. Normal control group, model group, western medicine group, Gout group high, medium and low concentration group, Gout group optimized high, medium and low concentration group, respectively, with saline, indometacin suspension, , Low concentration decoction, gout combination of high side, medium and low concentration of decoction 7 days, morning and evening 1 times. On the fourth day after gavage, urate solution of 25 g / mL (0.05 mL) was injected into rat ankle cavity to prepare gouty arthritis model. Rats in control group were injected with 0.05 mL saline. The rats were weighed before and 24 h, 48 h and 72 h after gavage, respectively. The rats were weighed in the morning and afternoon on the day before model making, and were sacrificed at 0,2,4,6,8,10,12,24 , 48,72h measured right leg from the same part of the circumference of the calf (mean); 72h after modeling, blood was collected to observe the plasma leukocyte count, serum levels of Interleukin-1β (IL-1β) The rats were sacrificed after taking blood, ankle joints were taken, and 10% formalin was fixed for pathology. Results: All 54 rats were involved in the result analysis. There was no difference in body weight before gavage. The gout group, high concentration group and western medicine group were higher than the model group on the 1st and 2nd day after modeling, there was no difference in the ankle circumference at morning and afternoon before modeling, The degree of swelling in rats after modeling was higher in all groups, especially in model group. After treatment, the degree of swelling in treatment group decreased significantly, while the effect in high concentration group and indomethacin was comparable. Compared with the model group, the levels of plasma leukocyte and serum interleukin-1βlevels in each dose group were significantly higher than those in the control group, and the difference was statistically significant between the Gout group and the optimized high-concentration group. The histopathological results showed that the Gout group And the optimized prescription can reduce the tissue edema and infiltration of inflammatory cells in the ankle joint of gouty joint rats and improve the synovial hyperplasia. The higher the concentration is, the more obvious the effect is. However, the above test indicators, gout combination group and optimization group no difference. Conclusion: Gout Mixture and Optimum Prescription can effectively inhibit the onset of acute gouty arthritis, and the dose-dependent manner, but the effect of Gout Mixture and optimization of the parties there is no difference.
