目的比较黄曲霉毒素B1(AFB1)与乙肝病毒(HBV)协同诱发树鼩肝细胞癌生长不同阶段的蛋白质表达谱的改变,筛查在AFB1与HBV共同诱发的肝癌发生发展中起重要作用的关键蛋白质分子。方法用双向凝胶电泳(2-DE)及基质辅助激光解吸飞行时间串联质谱法(MALDI-TOF-MS/MS)对同一动物自身的癌发生前的肝组织(C45w)、癌发生后的肝癌组织(CCa)及未经AFB1处理的对照组动物肝组织(E45w、E105w)进行了相互比较。结果 C45w组、CCa组、E45w、E105w组分别平均检测到(1233±30)个蛋白点(n=3),(1257±35)个蛋白点(n=3),(1249±51)个蛋白点(n=3)和(1208±36)个蛋白点(n=3)。以各组中2-DE效果好、点数多的一块胶作为参考胶,分别进行两组间的匹配分析(E45wvsC45w、C45wvsCCa、E45wvsE105w),筛选出两组之间差异大于或等于2倍的蛋白点65个。应用质谱鉴定出细胞色素C氧化酶等25种胶内差异蛋白质。结论在AFB1与HBV共同诱发的树鼩肝癌发生发展不同阶段有明显的差异性蛋白表达谱,为进一步对人肝癌蛋白质组的研究提供了线索。
OBJECTIVE: To compare the changes of protein expression profiles in different stages of hepatocellular carcinoma induced by aflatoxin B1 (AFB1) and hepatitis B virus (HBV), and to screen the key factors that play an important role in the carcinogenesis and development of hepatocellular carcinoma induced by AFB1 and HBV Protein molecules. Methods Two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser desorption / time of flight tandem mass spectrometry (MALDI-TOF-MS / MS) Liver tissues (E45w, E105w) from control (CCa) and non-AFB1 treated animals were compared with each other. Results The average number of (1233 ± 30) protein spots (n = 3), (1257 ± 35) protein spots (n = 3) and (1249 ± 51) spots were detected in C45w group, CCa group, E45w group and E105w group Points (n = 3) and (1208 ± 36) protein spots (n = 3). The 2-DE with good effect and the number of points in each group was used as a reference glue, and the matching analysis (E45wvsC45w, C45wvsCCa, E45wvsE105w) was conducted between the two groups, and the protein spots with the difference of two times or more were screened out 65. 25 kinds of differential proteins in the gel were identified by mass spectrometry, such as cytochrome C oxidase. Conclusions The differentially expressed protein profiles at different stages of the development of tree shrew hepatocellular carcinoma induced by AFB1 and HBV may provide distinct clues for the further study on the proteome of human liver cancer.
