目的探讨不同浓度的短管兔耳草乙醇提取物对H_(22)荷瘤小鼠肿瘤细胞生长及血清IL-2和TNF-ɑ的影响。方法采用细胞接种的方法建立小鼠肝癌H_(22)移植瘤模型,随机分为模型对照组,顺铂组,短管兔耳草乙醇提取物高、中、低剂量组,另设正常对照组。观察各组小鼠一般状况,检测有效体质量变化及抑瘤率、肿瘤指数、肝脏、脾脏、胸腺、和肾脏的器官指数,比色分析法检测肝组织中SOD和MDA的含有量,ELISA检测血清中IL-2和TNF-ɑ的含有量。结果短管兔耳草乙醇提取物各干预组的一般状况优于模型对照组和顺铂组;各治疗组瘤体质量均低于模型对照组瘤质量(P<0.05或P<0.01);短管兔耳草乙醇提取物各干预组有效体质量增加,胸脾指数高于模型对照组(P<0.05或P<0.01);同时,短管兔耳草乙醇提取物各干预组血清SOD的活性、IL-2和TNF-ɑ的含有量均高于模型对照组(P<0.05或P<0.01),血清MDA的含有量及肝肾指数低于模型组(P<0.05或P<0.01)。结论短管兔耳草乙醇提取物可抑制小鼠肝癌H_(22)肿瘤细胞的增殖,其机制可能与增强血清IL-2和TNF-ɑ的含有量有关,也可能与增强机体的抗氧化能力有关。
Objective To investigate the effects of different concentrations of ethanol extract from Tamarix ramosissima on tumor growth, serum IL-2 and TNF-ɑ in H22 tumor-bearing mice. Methods H22 hepatocellular carcinoma xenografts in mice were established by cell inoculation, and were randomly divided into model control group, cisplatin group, high and medium and low dose groups of ethanol extract of short tube of Eupatorium majus and normal control group . The mice in each group were observed for their general condition, the change of effective body mass, the tumor inhibition rate, tumor index, liver, spleen, thymus and kidney organ indexes. The content of SOD and MDA in liver tissue were detected by colorimetric assay. Serum IL-2 and TNF-含 content. Results The results showed that the ethanol extract of T. auriculae was better than the model control group and the cisplatin group in the intervention group. The tumor mass of each treatment group was lower than that of the model control group (P <0.05 or P <0.01) The effective body weight of the ethanol extract of Tamarix ramosissima increased in each intervention group, the thoracic spleen index was higher than the model control group (P <0.05 or P <0.01); at the same time, the activity of SOD (P <0.05 or P <0.01). The content of MDA and the index of liver and kidney in serum were lower than those in model group (P <0.05 or P <0.01). Conclusion The ethanol extract of T. obliqua can inhibit the proliferation of H22 tumor cells in mice, which may be related to the increase of serum levels of IL-2 and TNF-,. It may also be related to the enhancement of antioxidant capacity related.
