目的分析肿瘤转移抑制基因-1(TMSG-1)在前列腺癌细胞株中与前列腺癌组织中的表达及其临床意义。方法 PCR法检测TMSG-1在人不同转移潜能前列腺癌细胞株低转移潜能(PC-3M-284)和高转移潜能(PC-3M-IE8)中的表达,Matrigel穿膜实验检测肿瘤细胞体外侵袭能力,采用免疫组织化学方法检测TMSG-1在人前列腺增生及前列腺癌组织中的表达。结果两种不同转移潜能前列腺癌细胞株中TMSG-1 mRNA的相对表达量之间差异有统计学意义(P<0.05)。TMSG-1在低转移潜能(PC-3M-2B4)细胞株的mRNA(2.8±0.3)明显高于高转移潜能细胞株PG-IE8的表达(1.2±0.2)。免疫组化检测TMSG-1在前列腺增生阳性表达高于前列腺癌表达,差异显著;Matrigel穿膜侵袭实验结果显示,3个正义转染组的穿膜细胞明显减少,穿膜细胞数分别为42.31±3.16、52.00±1.83和23.93±2.79,与空白对照组(82.01±3.14)和空载体对照组(78.00±3.02)相比,均明显减少,差异具统计学意义(P<0.05)。结论 TMSG-1具有抑制前列腺癌细胞生长、促进其凋亡、抑制其侵袭能力等重要功能,TMSG-1的表达与肿瘤细胞转移潜能呈负相关。
Objective To analyze the expression of tumor metastasis suppressor gene-1 (TMSG-1) in prostate cancer cell lines and prostate cancer and its clinical significance. Methods The expression of TMSG-1 in low metastatic potential (PC-3M-284) and high metastatic potential (PC-3M-IE8) of human prostate cancer cell lines with different metastatic potential was detected by PCR. Matrigel transmembrane permeability The expression of TMSG-1 in human benign prostatic hyperplasia and prostate cancer tissues was detected by immunohistochemistry. Results The relative expression of TMSG-1 mRNA in two different metastatic potential prostate cancer cell lines was significantly different (P <0.05). TMSG-1 mRNA (2.8 ± 0.3) in low metastatic potential (PC-3M-2B4) cell line was significantly higher than that in high metastatic potential cell line PG-IE8 (1.2 ± 0.2). The positive rate of TMSG-1 in benign prostatic hyperplasia was higher than that in prostatic adenocarcinoma by immunohistochemistry, and the difference was significant. The transmembrane invasion assay of Matrigel showed that the transmembrane cells in the three positive transfection groups were significantly decreased, the number of transmembrane cells were 42.31 ± 3.16,52.00 ± 1.83 and 23.93 ± 2.79, respectively. Compared with the blank control group (82.01 ± 3.14) and the empty vector control group (78.00 ± 3.02), the levels were significantly decreased, with statistical significance (P <0.05). Conclusion TMSG-1 has the important function of inhibiting the growth of prostate cancer cells, promoting its apoptosis and inhibiting its invasion ability. The expression of TMSG-1 is negatively correlated with the metastasis potential of tumor cells.
