目的:研究彩色多普勒超声辐照中孕大鼠,对胎鼠肾脏P53蛋白表达的影响。方法:彩超辐照孕14dSD大鼠子宫体表投影区,按照射时间不同分为对照组、照射10min组、20min组、30min组。仪器采用Acuson公司Sequoia-512型彩色电脑声像仪,照射条件:4V1探头,二维频率H3.0MHz,彩色频率3.0MHz,热指数(TI)=1.8,机械指数(MI)=1.6。各组于既定照射结束后24h取胎鼠肾脏标本,HE染色光镜下观察肾脏组织结构,免疫组化方法检测P53蛋白的表达。结果采用统计软件SPSS11.0行多组均数比较的方差分析。结果:光镜观察各组胎鼠肾脏组织均未见明显损伤。免疫组化法检测,各组胎鼠肾小管上皮细胞内可见均匀散在的胞核棕黄色淡染的P53蛋白阳性表达。照射10min、20min组P53蛋白表达与对照组无显著差异(P>0.05);照射30min组P53蛋白表达增强,与对照组有显著差异(P<0.05)。结论:彩超辐照中孕期SD大鼠子宫投影区30min内,不会引起胎鼠肾脏明显的组织学损伤;照射30min,胎鼠肾脏P53蛋白表达增加。
Objective: To study the effect of color Doppler ultrasound irradiation on the expression of P53 protein in fetal rat kidney. Methods: The uterine projection area of 14 d SD rats irradiated by color Doppler ultrasound was divided into control group, irradiated 10 min group, 20 min group and 30 min group. The instrument uses Acuson Sequoia-512 color computer imager, irradiation conditions: 4V1 probe, two-dimensional frequency H3.0MHz, color frequency of 3.0MHz, heat index (TI) = 1.8, mechanical index (MI) = 1.6. The kidneys of fetus were harvested at 24h after irradiation. The structure of kidney was observed by HE staining and the expression of P53 protein was detected by immunohistochemistry. Results using statistical software SPSS11.0 multiple groups of means of variance analysis. Results: There was no obvious damage in the kidneys of each group. Immunohistochemical method was used to detect the positive expression of P53 protein in the light brown stained cells scattered evenly in the renal tubular epithelial cells in each group. The expression of P53 protein in 10 min and 20 min groups was not significantly different from that in the control group (P> 0.05). The P53 protein expression in the 30 min irradiation group was significantly increased (P <0.05). Conclusion: Within 30min of the uterine projection area of SD rats during the color Doppler ultrasound irradiation, obvious histological damage will not occur in the fetal rat kidneys. After irradiation for 30min, the expression of P53 protein in fetal rat kidneys increased.
