目的:采用响应面法对北青龙衣总多糖的提取工艺进行优化。方法:以北青龙衣总多糖提取量为指标,在单因素试验基础上,选择提取温度、料液比、提取时间为考察因素,根据Box-Behnken实验设计原理,利用响应面法优化北青龙衣中总多糖的提取工艺。结果:最佳提取工艺为提取温度88℃,料液比1∶22,提取时间72min,多糖提取量实测值7.83 mg·g-1,与预测值8.02 mg·g-1较为接近。结论:采用响应面法优选北青龙衣总多糖提取工艺稳定可行,为北青龙衣资源开发利用及总多糖提取工艺产业化提供理论依据。
Objective: To optimize the extraction process of total polysaccharide of Beilong Qingyi by response surface methodology. Methods: Based on the single factor experiment, the extraction temperature, the ratio of solid to liquid and the extraction time were selected as the indexes by taking the total polysaccharide extract of Beilong Qingyi as an index. According to the Box-Behnken experimental design principle, the response surface methodology was used to optimize Beilong drape The total polysaccharide extraction process. Results: The optimum extraction conditions were: extraction temperature 88 ℃, solid-liquid ratio 1:22 and extraction time 72 min. The measured value of polysaccharide extraction was 7.83 mg · g-1, which was close to the predicted value of 8.02 mg · g-1. Conclusion: The optimization of the extraction process of total polysaccharides from Beilong Qingyi by response surface methodology is stable and feasible, which provides a theoretical basis for the exploitation and utilization of Beilong Qingyi resources and the industrialization of total polysaccharide extraction process.
