固本防哮饮对哮喘缓解期小鼠肺组织IL-4及IL-12的影响

目的:研究固本防哮饮对哮喘缓解期小鼠肺组织白介素4(IL-4)、白介素12(IL-12)的干预作用,探讨其治疗哮喘的免疫学机制。方法:75只小鼠随机分为正常组、模型组、预防组、固本防哮饮组和孟鲁司特钠组,以卵蛋白(OVA)和呼吸道合胞病毒(RSV)联合致敏激发制备哮喘缓解期小鼠模型。观察各组小鼠的一般行为活动和病理学变化特点,用RT-PCR法测定哮喘小鼠肺组织IL-4、IL-12的m RNA表达。结果:模型组小鼠肺组织可见支气管壁水肿、增厚,周围有嗜酸性粒细胞和中性粒细胞浸润。预防组、固本防哮饮组及孟鲁司特钠组,小鼠肺病理改变显著改善,支气管壁充血水肿现象减轻,管壁和平滑肌厚度减小,且嗜酸性粒细胞和中性粒细胞浸润也显著减少。模型组小鼠肺组织IL-4 m RNA较正常组显著高表达(P<0.05),而IL-12 m RNA较正常组低表达(P<0.05)。预防组、固本防哮饮组、孟鲁司特钠组IL-4 m RNA较模型组表达下降(P<0.05),而IL-12 m RNA较模型组表达升高(P<0.05)。结论:固本防哮饮防治哮喘的机制可能是通过降低IL-4 m RNA同时升高IL-12 m RNA表达来纠正Th1/Th2失衡,从而减轻气道炎性反应,缓解气道重塑。 Objective: To study the intervention effect of Guben anti-asthmatic beverage on interleukin-4 (IL-4) and interleukin-12 (IL-12) in the lung tissue of asthmatic mice during remission period and to explore its immunological mechanism of treating asthma. IL-4 mRNA expression in lung tissue of model group was significantly higher than that of normal group (P <0.05), while IL-12 mRNA was lower than that of normal group (P <0.05). The levels of IL-4mRNA in prevention group, Guben anti-asthma drink group and montelukast sodium group were lower than those in model group (P <0.05), while the expression of IL-12 mRNA in model group was higher than that in model group (P <0.05). Conclusion: The mechanism of Guben anti-asthma drink in preventing and treating asthma may be to correct the imbalance of Th1 / Th2 by decreasing IL-4 m RNA and IL-12 m RNA expression, thereby reducing airway inflammatory reaction and relieving airway remodeling.
