应用流式细胞术定量检测10例正常大肠粘膜、27例大肠腺瘤及32例大肠腺癌的p21~(ras)及DNA含量,结果发现:①正常大肠粘膜、大肠腺瘤及大肠癌的p21~(ras)表达量依次递增,并随大肠腺瘤不典型增生程度的加重以及瘤体的增大而增多;②大肠腺瘤组p21~(ras)表达增强呈阳性者的DI值、SPF值显著高于p21~(ras)阴性者,而大肠癌组仅SPF值有显著性差异;③DNA含量呈异倍体的大肠腺瘤及腺癌的p21~(ras)表达量显著高于DNA二倍体者本研究从p21~(ras)表达的量变意义上说明:①p21~(ras)过表达促进了大肠肿瘤细胞的异常增殖,并导致细胞DNA含量异常,出现DNA异倍体;②p21~(ras)表达量的变化在大肠腺瘤向腺癌恶性转化的质变过程中可能具有重要作用。
Flow cytometry was used to quantitatively detect the p21 ras and DNA contents in 10 normal colorectal mucosa, 27 colorectal adenomas and 32 colorectal adenocarcinomas. The results showed that: 1 Normal colorectal mucosa, colorectal adenoma, and colorectal cancer p21 The expression of ras increased progressively, and increased with the aggravation of the atypical hyperplasia of colorectal adenomas and the increase of tumor size. 2 The DI value and SPF value of the positive expression of p21 ras in colorectal adenomas Significantly higher than those with negative p21 ras, while the colorectal cancer group had only significant difference in SPF value; 3 The expression of p21 ras in colorectal adenoma and adenocarcinoma with aneuploid DNA was significantly higher than that in DNA. In this study, from the quantitative change of p21 ras expression, 1p21 ras overexpression promoted the abnormal proliferation of colorectal tumor cells, and resulted in abnormal DNA content and DNA aneuploidy; 2p21~(ras) ) Changes in expression levels may play an important role in the process of malignant transformation of colorectal adenomas to adenocarcinoma.
