目的 :染料木素是大豆中含量最丰富的异黄酮之一 ,被认为可以预防绝经后骨质疏松。本研究从骨的合成代谢的角度探讨染料木素影响骨代谢的机理。方法 :用不同浓度的染料木素干预人成骨肉瘤细胞株SaOS2 ,观察对其增殖和分化功能的影响。用美蓝染色后测定细胞的吸光度的方法反映细胞增殖情况 ,用硷性磷酸酶活性反映细胞的分化程度。结果 :低浓度 10 -10 ~ 10 -8mol L的染料木素有促进细胞增殖的作用 (P <0 .0 1或 0 .0 5) ,但是在此浓度范围内染料木素抑制经蛋白校正的硷性磷酸酶的活性 (P <0 .0 1或 0 .0 5) ,从整体上讲 (不用蛋白校正 ) ,染料木素对硷性磷酸酶的活性无影响 (F =1.6 4 3,P >0 .0 5)。结论 :染料木素对骨的合成代谢无影响 ,其对骨质疏松的预防作用可能是通过其他的机制。
Purpose : Genistein is one of the most abundant isoflavones in soybean and is believed to prevent postmenopausal osteoporosis. This study explored the mechanism of genistein on bone metabolism from the perspective of bone anabolism. METHODS: Human osteogenic sarcoma cell line SaOS2 was treated with different concentrations of genistein to observe its effects on proliferation and differentiation. The method of measuring the absorbance of cells after staining with methylene blue reflects the cell proliferation, and the activity of alkaline phosphatase reflects the degree of cell differentiation. RESULTS: Genistein at a low concentration of 10 -10 to 10 -8 mol L promoted cell proliferation (P 0.01 or 0.05), but genistein suppressed protein-corrected concentrations in this concentration range. Activity of alkaline phosphatase (P <0. 01 or 0. 05), as a whole (without protein correction), genistein had no effect on the activity of alkaline phosphatase (F = 1.6 4 3, P > 0 .0 5). Conclusion : Genistein has no effect on bone anabolism. Its preventive effect on osteoporosis may be through other mechanisms.
