目的观察单克隆天然抗角蛋白抗体384与白念珠菌结合及对芽管生成的影响。方法采用ELISA和流式细胞仪检测单克隆天然抗角蛋白抗体384与白念珠菌结合,并将抗体与真菌共孵育,检测白念珠菌芽管生成的变化。结果ELISA和流式细胞仪检测均发现3B4可以与白念珠菌结合;相对于同型对照组,384作用组真菌芽管生成率显著下降,两组差异有统计学意义。结论单克隆天然抗角蛋白抗体3B4可以与白念珠菌结合并抑制其芽管生成,提示天然IgM在机体抗真菌天然免疫中可能具有一定的作用。
Objective To observe the effect of monoclonal anti-keratin antibody 384 on Candida albicans and on the formation of germ tube. Methods Monoclonal anti-keratin antibody 384 was bound to Candida albicans by ELISA and flow cytometry. The antibody and fungi were co-incubated to detect the changes of Candida albicans germ tube formation. Results ELISA and flow cytometry showed that 3B4 could bind to Candida albicans. Compared with the control group, the formation rate of fungus germ tube in 384 treatment group was significantly decreased, with significant difference between the two groups. Conclusion Monoclonal natural anti-keratin antibody 3B4 can bind to Candida albicans and inhibit its germ tube formation, suggesting that native IgM may play a role in the natural antifungal immunity.
