气相色谱(GC)是测定鱼类脂肪 W-3系多不饱和脂肪酸的常用方法,一般是先将生物样本中的脂肪提取并甲酯化,然后用 GC 法测定。但是,海产油脂的脂肪酸种类很多,极其复杂,需要在最佳色谱条件下加以区分以免谱峰重叠。在甲酯化过程中,随着甲酯产物的形成还会带来另外的问题,因此,最终需用 GC 一群色谱测定,质子核磁共振谱(H—NMR)是一种全新的快速测定 W-3多不饱和脂肪酸结构的方法,它是利用 W-3系多不饱和脂肪酸(δ=0.95PPM)中的甲基共振现象以及共振产生的化学位移来进行测定
Gas chromatography (GC) is a common method for the determination of W-3 polyunsaturated fatty acids in fish fats. The fats in biological samples are first extracted and methyl esterified and then measured by GC. However, there are many types of fatty acids in seafood oils that are extremely complex and need to be distinguished under the best chromatographic conditions in order to avoid overlapping peaks. In the methyl esterification process, additional problems arise with the formation of the methyl ester product. Therefore, a group of GC chromatograms are finally required. Proton nuclear magnetic resonance spectroscopy (H-NMR) is a new and rapid method for the determination of W- 3 polyunsaturated fatty acid structure by the methyl resonance phenomenon in the W-3 polyunsaturated fatty acid (δ = 0.95PPM) and the chemical shift due to resonance
