目的 :观察缓慢牵伸肢体延长时周围神经相应的脊髓及背根神经节(DRG)中生长相关蛋白 43(GAP 43)mRNA表达。方法 :29只日本大耳白兔胫骨以1mm/d的速度缓慢牵伸延长至20 %、40 %,停止延长后2、4周 ,用原位杂交技术观察坐骨神经相应背根神经节及脊髓中GAP 43mRNA的表达及定位 ,以图像分析仪进行半定量分析。结果 :缓慢牵伸至20 %时相应脊髓及神经节呈阳性表达 ,延长40 %时呈强阳性表达 ,停止延长4周恢复正常。结论 :缓慢牵伸周围神经虽无神经临床损害症状 ,但引起GAP 43mRNA表达的上调 ,并在停止延长后4周恢复正常 ;缓慢牵伸过程中神经的亚临床损害广泛存在 ,并存在着强烈的神经再生反应。
OBJECTIVE: To observe the mRNA expression of growth-associated protein 43 (GAP 43) in the spinal cord and dorsal root ganglion (DRG) of the peripheral nerve when the limb is extended slowly. Methods: Twenty-nine Japanese white rabbits were slowly drawn and extended to 20% and 40% at a speed of 1 mm / d, respectively. After 2 and 4 weeks of extension, the corresponding dorsal root ganglia and spinal cord of the sciatic nerve were observed by in situ hybridization GAP 43 mRNA expression and localization, semi-quantitative analysis with an image analyzer. Results: Slowly drafted to 20% of the corresponding spinal cord and ganglion-positive expression was 40% extended strong positive expression, stop extending to 4 weeks returned to normal. CONCLUSION: Although there is no clinical symptom of nerve injury in slow draining peripheral nerves, GAP 43 mRNA expression is up-regulated and returned to normal after 4 weeks of prolonged withdrawal. The subclinical nerve damage during slow drafting is widespread and there is a strong Nerve regeneration reaction.
