目的探讨促凋亡基因PDCD5 mRNA在初诊Graves病(GD)和桥本甲状腺炎(HT)患者外周血单个核细胞(PBMC)中的表达。方法半定量RT-PCR法测定50例GD和30例HT患者PBMC中PDCD5 mRNA表达的阳性率和表达水平。结果GD和HT患者PBMC中PDCD5 mRNA表达的阳性率和表达水平均高于正常对照组,其中HT患者PDCD5 mRNA表达的阳性率显著高于正常对照组(P<0.05),GD、HT、正常对照三组间PDCD5 mRNA表达水平差异有统计学意义(P<0.01),HT组PDCD5 mRNA表达水平显著高于GD组(P<0.01)。患者PDCD5 mRNA表达水平与甲状腺微粒体抗体(TM-Ab)、甲状腺球蛋白抗体(TG-Ab)呈显著正相关(P<0.01),与血清T4水平及|lgTSH|呈显著负相关(P<0.05)。结论PDCD5 mRNA在GD和HT患者PBMC中表达升高,可能与GD和HT淋巴细胞凋亡异常有关。
Objective To investigate the expression of PDCD5 mRNA in peripheral blood mononuclear cells (PBMCs) of patients with newly diagnosed Graves disease (GD) and Hashimoto’s thyroiditis (HT). Methods The positive rate and expression level of PDCD5 mRNA in peripheral blood mononuclear cells from 50 GD patients and 30 HT patients were determined by semi-quantitative RT-PCR. Results The positive rate and expression level of PDCD5 mRNA in peripheral blood mononuclear cells of GD and HT patients were higher than those of normal controls. The positive rate of PDCD5 mRNA in HT patients was significantly higher than that in normal controls (P <0.05), GD, HT, normal controls The expression of PDCD5 mRNA in three groups had statistical significance (P <0.01). The expression of PDCD5 mRNA in HT group was significantly higher than that in GD group (P <0.01). There was a significant positive correlation between the expression of PDCD5 mRNA and thyroid microsomal antibody (TM-Ab) and thyroglobulin antibody (TG-Ab) (P <0.01) and the serum T4 level and |¤g TSH | 0.05). Conclusion The expression of PDCD5 mRNA in PBMC of GD and HT patients is elevated, which may be related to the abnormal apoptosis of GD and HT lymphocytes.
