从本实验室分离得到的 4 0 6株苏云金芽孢杆菌中 ,以甜菜夜蛾为供试昆虫 ,采用生物测定的方法 ,筛选出一株对甜菜夜蛾高效的WY 1 90菌株 ,确定其发酵原液LC5 0 为 0 .2 5μl/ml。对WY 1 90的形态特征、生理生化特性和伴孢晶体蛋白进行了观察和研究。研究结果表明 ,WY 1 90属于H3a3b血清型库斯塔克亚种 ;具有几丁质酶活性 ,伴孢晶体形态主要为大小不一的菱形、小正方形、椭圆形 ,并具镶嵌结构 ;SDS PAGE电泳图谱表明主要由 1 35kD和 6 5kD两种蛋白成分组成 ,在菌株生长形态、发酵培养特征、生理生化特性方面与生产菌株HD 1的差别不大 ,其主要差异表现在对甜菜夜蛾的敏感性和杀虫毒力的差别上。
A total of 460 strains of Bacillus thuringiensis isolated from this laboratory were tested for their high bioavailability to beet armyworm Spodoptera exigua by using the bioassay method. LC5 0 is 0.5 2 μl / ml. The morphological, physiological and biochemical characteristics of WY 1 90 and its crystal protein were observed and studied. The results showed that WY 1 90 belongs to the genus H3a3b serotype Crestak, and has the chitinase activity. The crystal forms of the sporozoite mainly consist of rhombus, small square, oval and inlaid structures with different sizes. SDS PAGE Electrophoregram showed that the two proteins were mainly composed of 35kD and 65kD protein components. There was no significant difference in growth morphology, fermentation characteristics and physiological and biochemical characteristics between strain HD 1 and strain HD 1. The main difference was the sensitivity to beet armyworm Differences in sex and insecticidal virulence.
