以不同分子质量组分的马尾藻岩藻聚糖(SF)为研究对象,体外培养小鼠肉瘤细胞S-180,做抗肿瘤活性试验。采用MTT法测定不同铺板数量和不同浓度SF作用下的细胞生长曲线,确定细胞的铺板数量和SF对肿瘤细胞增殖抑制作用的有效浓度范围;测定不同组分SF在不同浓度、时间条件下对该细胞的增殖抑制率及对肿瘤细胞的IC50值。在倒置显微镜下观察SF2-R对细胞形态的影响。试验结果显示,当细胞铺板数量为5×103个/mL时,由细胞数量反映的OD值与时间的线性相关性良好;当SF的质量浓度设定在1~25 mg/mL范围时,细胞增殖抑制作用与SF浓度线性相关性良好;8种分子质量组分的SF对肿瘤细胞均有抑制作用,其中SF2-R对细胞的抑制作用最强。SF2-R对S-180肿瘤细胞的IC50为7.88 mg/mL,低于其他组分对肿瘤的IC50。倒置显微镜观察结果是:随着SF浓度的增加,细胞凋亡加重。
Using different molecular weight components of Sargassum fusatum polysaccharide (SF) as the research object, the mouse sarcoma S-180 cells were cultured in vitro and the anti-tumor activity was tested. The IC50 of SF2-R on S-180 tumor cells was 7.88 mg / mL, which was lower than the IC50 of other components on tumors. Inverted microscope observation results showed that as SF concentration increased, apoptosis increased.
