目的通过分析不同病变组织表达癌胚抗原信使核糖核酸 ( CEA-m RNA)的差异 ,探讨 CEA-m RNA指标诊断消化道恶性肿瘤的应用价值。方法以 35例消化道恶性肿瘤患者和 12例非恶性肿瘤的良性消化道疾病患者为研究对象 ,收集新鲜肿瘤组织及其癌旁正常组织和非恶性肿瘤组织以及同体血液为检测标本 ,应用套式反转录聚合酶链反应 ( RT-PCR)检测组织标本中 CEA-m RNA、酶联免疫吸附试验 ( EL ISA)检测血液标本癌胚抗原 ( CEA)。结果恶性消化道肿物患者瘤组织标本 2 9例 ( 82 .9% )表达 CEA-m RNA,血液标本 15例 ( 42 .9% )表达 CEA-m RNA,两个指标表达情况相比具有显著性差异 ( P<0 .0 1)。癌旁正常组织非癌病理组织也分别检测 4例和 1例表达 CEA-m RNA阳性结果 ,组织标本 CEA-m RNA指标诊断恶性消化道肿瘤的特异性为 95 .0 %、敏感性为 82 .9%。结论在诊断消化道恶性肿瘤方面 ,CEA-m RNA是特异性和敏感性较好且很实用的指标之一 ,可以常规应用
Objective To investigate the value of CEA-m RNA in the diagnosis of gastrointestinal malignancies by analyzing the difference of carcinoembryonic antigen messenger ribonucleic acid (CEA-m RNA) expression in different pathological tissues. Methods Twenty-five patients with gastrointestinal cancer and 12 patients with benign gastrointestinal diseases who were non-malignant were enrolled in this study. Fresh tumor tissue, normal tissue adjacent to tumor and non-malignant tumor tissue were collected. CEA-m RNA was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) in tissue samples, and CEA was detected by enzyme linked immunosorbent assay (ELISA). Results The CEA-m RNA was detected in 29 cases (82.9%) of malignant gastrointestinal masses and CEA-m RNA was expressed in 15 cases (42.9%) of the blood samples. There were significant differences between the two indicators Sex differences (P <0.01). The positive expression of CEA-m RNA was also detected in 4 cases and 1 case of normal non-cancerous tissues respectively. The specificity of CEA-m RNA in diagnosis of malignant gastrointestinal tumors was 95.0% and the sensitivity was 82%. 9%. Conclusion In the diagnosis of gastrointestinal cancer, CEA-m RNA is one of the more specific and sensitive indicators and can be applied routinely
