目的探讨肝素对HSC生长、细胞外基质和基质金属蛋白酶基因表达的影响。方法用肝素和不用肝素对活化的HSC进行处理。以直接细胞计数和BrdU标记免疫细胞化学染色检测细胞的生长情况,用免疫细胞化学染色和细胞原位核酸分子杂交分别检测Ⅰ、Ⅳ型前胶原蛋白、纤连蛋白和Ⅰ、Ⅳ型胶原、纤连蛋白、基质金属蛋白酶-2及膜型基质金属蛋白酶基因的表达,并用酶图法检测基质金属蛋白酶-2的活性。结果肝素使血清所致的HSC生长速度减慢;肝素抑制HSCⅠ型前胶原、纤连蛋白的合成和Ⅰ型前胶原、纤连蛋白、膜型基质金属蛋白酶基因的表达,但对Ⅳ型胶原、基质金属蛋白酶-2 mRNA水平和基质金属蛋白酶-2活性无影响。结论肝素明显抑制血清所致的HSC的生长;下调HSC间质型胶原和FN的合成,并抑制膜型基质金属蛋白酶基因的表达。
Objective To investigate the effect of heparin on HSC growth, extracellular matrix and matrix metalloproteinase gene expression. Methods Activated HSCs were treated with heparin and without heparin. The cell growth was detected by direct cell counting and BrdU labeling immunocytochemistry. The expressions of type Ⅰ and Ⅳ procollagen, fibronectin, type Ⅰ and Ⅳ collagen, fibronectin and fibronectin were detected by immunocytochemistry and in situ hybridization The expression of connexin, MMP - 2 and MMP - 2 were detected by enzyme - linked immunosorbent assay (ELISA). Results Heparin decreased the serum HSC growth rate. Heparin inhibited the expression of HSC type Ⅰ procollagen and fibronectin, and the expression of procollagen type Ⅰ, fibronectin and membrane type matrix metalloproteinases. However, Matrix metalloproteinase-2 mRNA level and matrix metalloproteinase-2 activity had no effect. Conclusion Heparin can significantly inhibit the growth of serum-induced HSC, down-regulate the synthesis of interstitial collagen and FN, and inhibit the expression of membrane-type matrix metalloproteinase gene.
