目的:建立藤梨根药材中熊果酸、齐墩果酸的含量测定方法,对不同品种、产地药材进行测定。方法:采用反向高效液相色谱法,色谱柱为ZORBA×300SB-C18(4.6 mm×250 mm,5μm),流动相甲醇-0.5%醋酸胺(76∶24),流速为1.0 mL.min-1,柱温25℃,检测波长210 nm。结果:熊果酸在0.492~2.460μg线性关系良好(r=0.999 7),平均回收率为97.6%,RSD 1.8%;齐墩果酸在0.448~2.240μg线性关系良好(r=0.999 9),平均回收率为98.7%,RSD 0.6%。结论:该方法简单、准确并且专属性强,可作为藤梨根药材质量控制方法。
OBJECTIVE: To establish a method for the determination of ursolic acid and oleanolic acid in Tengligen medicinal materials, and to determine the content of ursolic acid and oleanolic acid in different varieties and producing areas. Methods: Reverse phase high performance liquid chromatography (HPLC) was used. The mobile phase consisted of ZORBA × 300SB-C18 (4.6 mm × 250 mm, 5 μm), mobile phase methanol- 1, column temperature 25 ℃, detection wavelength 210 nm. Results: Ursolic acid showed a good linearity (r = 0.999 7) at a linear range of 0.492-2.460 μg with an average recovery of 97.6% and a RSD of 1.8%. Oleanolic acid had a good linear relationship at 0.448-2.240 μg (r = 0.999 9) The average recovery was 98.7% with a RSD of 0.6%. Conclusion: The method is simple, accurate and specific and can be used as a quality control method for Tengligen.
