目的:观察大黄脾虚模型大鼠壁细胞内CaM及CaCa2+/CaM-PK II活性的变化以及补脾方药党参、白术、补中益气汤对其的作用。方法:EDTA-Pronase酶消化法及Perco ll密度梯度离心分离和纯化大鼠胃壁细胞,根据的CaM的环核苷酸磷酸二酯酶检测法(PDE酶法)检测大鼠壁细胞内CaM活性;根据CaCa2+/CaM-PK II活性与激活底物磷酸化量成线性关系,测定其活性。结果:大黄脾虚模型大鼠壁细胞内CaM及CaCa2+/CaM-PKII活性均明显升高,补脾方药对其均有明显下调作用。结论:脾虚大鼠壁细胞处于高应激状态,党参、白术、补中益气汤等补脾方药可因对细胞内受体后信号传导物质的影响而调节细胞功能。
Objective: To observe the changes of CaM and CaCa2+/CaM-PK II activity in parietal cells of rhubarb spleen deficiency model rats and the effects of Codonopsis pilosula, Atractylodes rhizome and Buzhong Yiqi Decoction on them. METHODS: Rat gastric parietal cells were isolated and purified by EDTA-Pronase enzyme digestion and Perco ll density gradient centrifugation. CaM activity was measured in rat parietal cells according to the CaM cyclic nucleotide phosphodiesterase assay (PDE enzyme method). Based on the linear relationship between CaCa2+/CaM-PK II activity and the amount of activated substrate phosphorylation, the activity was determined. Results: The activities of CaM and CaCa2+/CaM-PKII in parietal cells of rats with spleen deficiency syndrome were all significantly increased. Conclusion: The parietal cells of rats with spleen deficiency are in high stress state. The functions of Dangshen, Atractylodes, Buzhong Yiqi Decoction and other recipes for supplementing spleen can regulate the function of cells due to the influence of signal transduction substances in receptors in cells.
