目的 :探讨电磁脉冲 (EMP)辐射对离体培养兔角膜上皮细胞的损伤作用。方法 :6× 10 4V/m场强的EMP重复照射离体培养的角膜上皮细胞 5次 ,应用原子力显微镜和生化检测仪 ,对辐射前后的细胞膜状态及上清液多种离子浓度进行检测。结果 :EMP照射后角膜上皮细胞膜表面出现多处大小不等的凹陷和穿孔。培养细胞上清多种离子浓度在照射后不同时间均有明显变化 ,其中K+ ,Ca2 + ,Fe2 + 浓度在照射后 2 4h明显升高。结论 :细胞膜是EMP作用于细胞的敏感靶部位 ,细胞膜穿孔是EMP非热效应的损伤作用机制之一。
Objective: To investigate the damage of cultured rabbit corneal epithelial cells by electromagnetic pulse (EMP) radiation. Methods: The corneal epithelial cells cultured in vitro were irradiated with 6 × 10 4 V / m EMP repeatedly for 5 times. Atomic force microscope and biochemical detector were used to detect the cell membrane state before and after irradiation and various ion concentrations of supernatant. Results: There were many dimples and perforations in corneal epithelial cell membrane surface after EMP irradiation. The concentrations of K +, Ca2 + and Fe2 + in the supernatant of cultured cells varied significantly at different times after irradiation. The concentrations of K +, Ca2 + and Fe2 + were significantly increased at 24 hours after irradiation. CONCLUSION: Cell membrane is a sensitive target of EMP on cells. Cell membrane perforation is one of the mechanisms of EMP non-thermal effects.
