目的:研究胍丁胺对氧糖剥夺/复氧损伤后 U251细胞水通道蛋-4(AQP4)和水通道蛋白-9(AQP9)表达的干预作用。方法氧糖剥夺/复氧损伤U251细胞后,在倒置显微镜下观察胍丁胺干预后U251细胞的形态变化, MTT法检测细胞存活率,Western blotting法分析AQP4和AQP9的表达。结果胍丁胺改善了缺血再灌注损伤后U251细胞的形态,显著提高了细胞存活率,明显抑制了缺血再灌注损伤后AQP4和AQP9的过度表达。结论胍丁胺通过抑制AQP4和AQP9的过度表达,减轻缺血再灌注损伤后继发的脑水肿,从而对神经元起到保护的作用。“,”Objective To investigate the effect of agmatine on aqua-porin 4 ( AQP4) and aquaporin 9 ( AQP9 ) expression after oxygen glu-cose deprivation-reperfusion injury in U251 cells.Methods The mor-phologic change of U251 cells was observed by inverted microscope.The viability of U251 cells were examined by MTT assay.Also, the expres-sion of AQP4 and AQP9 were detected by western blot analysis.Results After agmatine treatment, the morphous of U251 cells were improved and the viability of U251 cells were obviously increased.While the expression of AQP4 and AQP9 were decreased by agmatine treatment.Conclusion The protective effect of agmatine on neuron from ischemia-reperfusion injury is related to the inhibitory of AQP4 and AQP9 overexpression, then the brain edema can be relieved by the agma-tine treatment.
