[目的]探讨ABCC2在胆管细胞癌中的表达水平及其临床意义。[方法]收集湖北中医药大学附属医院病理科2010~2015年20例胆管细胞癌组织及其配对的10例远癌组织,利用实时荧光定量PCR和Western blot方法检测两组ABCC2的表达,比较分析ABCC2表达水平与胆管细胞癌的临床分期以及淋巴结转移的关系。[结果]ABCC2基因或蛋白在胆管细胞癌组织和远癌组织中均有不同程度的表达,且在癌组织的表达量显著低于远癌组织(P<0.05);此外,ABCC2表达在胆管细胞癌的不同淋巴结转移之间没有显著差异。[结论]ABCC2在胆管细胞癌中低表达(mRNA表达水平P=0.003,蛋白表达水平P=0.0031),且与淋巴结转移之间没有显著差异,可能在胆管细胞癌的发生发展中起一定作用,有助于胆管细胞癌的临床诊断和病情评估。
[Objective] To investigate the expression of ABCC2 in cholangiocarcinoma and its clinical significance. [Methods] Twenty cases of cholangiocarcinoma tissues and 10 paired cancerous tissues from 2010 to 2015 in Department of Pathology, Affiliated Hospital of Hubei University of Traditional Chinese Medicine were collected. The expression of ABCC2 in both groups was detected by real-time fluorescence quantitative PCR and Western blot. ABCC2 expression in cholangiocarcinoma clinical stages and lymph node metastasis. [Results] The expression of ABCC2 gene or protein in cholangiocarcinoma and distant tissues was significantly different, and the expression level in ABCA2 was significantly lower than that in distant carcinoma (P <0.05). In addition, the expression of ABCC2 in cholangiocarcinoma There was no significant difference between different lymph node metastases of cancer. [Conclusion] The low expression of ABCC2 in cholangiocarcinoma (mRNA expression level P = 0.003, protein expression level P = 0.0031) and there is no significant difference between lymph node metastasis and ABCC2 may play a role in the occurrence and development of cholangiocarcinoma, Contribute to the clinical diagnosis and assessment of cholangiocarcinoma.
