根据已知物种NBS抗病类基因(RGAs)保守序列设计引物,从芒果品种金煌基因组DNA中分离得到了10条同源序列(pp-1~10,GenBank登录号为HM446507~16)。DNA序列分析表明,这些RGAs在200~300 bp区间存在较大变异,Pi值都在0.4以上。同源性分析表明这些序列的同源性差异范围从11.0%~98.4%,离散值范围为1.6~100.7,10条RGAs可以分为2大类。蛋白序列分析表明,pp-01~10都具有开放读码框,编码的蛋白含有典型的NBS抗病类基因所拥有的P-loop和Kinase-2a结构域,通过同源进化分析可将其分为TIR-NBS-LRR和CC-NBS-LRR 2类,与已知物种同源性范围为20%~60.2%。
Ten homologous sequences (pp-1 ~ 10, GenBank accession number HM446507 ~ 16) were isolated from Jinhuang genomic DNA of mango cultivars according to the conservative sequences of known disease resistance genes (RGAs) of known species. DNA sequence analysis showed that these RGAs showed large variations in the range of 200-300 bp with Pi values above 0.4. Homology analysis showed that the homologies of these sequences ranged from 11.0% to 98.4% with discrete values ranged from 1.6 to 100.7, and 10 RGAs could be divided into two broad categories. Protein sequence analysis showed that pp-01-10 had an open reading frame, and the encoded protein contained the P-loop and Kinase-2a domains possessed by the typical NBS-resistant genes. By homologous evolutionary analysis, It is TIR-NBS-LRR and CC-NBS-LRR Class 2 with homology ranges 20% -60.2% with known species.
