目的观察散癖平胃(SPPW)方对人胃癌细胞SGC-7901侵袭转移能力的影响。方法 SPPW方含药血清干预人胃癌细胞SGC-7901 48h后,应用黏附实验、侵袭实验及迁移实验,检测SPPW方对人胃癌细胞SGC-7901侵袭转移能力的影响。应用实时定量聚合酶链反应(real-time PCR)检测SPPW对与侵袭转移有关的MMP-2和MMP-9mRNA表达的影响。结果 SPPW方各组含药血清干预人胃癌细胞48h后,SGC-7901细胞黏附抑制率、侵袭抑制率及迁移抑制率均增加且呈剂量依赖关系,与阴性对照组比较有统计学差异(P<0.01或P<0.05)。SPPW方三个剂量组对MMP-2mRNA的表达无统计学差异,对MMP-9mRNA的表达具有统计学差异(P<0.01)。结论 SPPW方可抑制人胃癌细胞SGC-7901黏附、侵袭和迁移能力,其抑制有明显的剂量依赖性,其抑制的机制可能与下调MMP-9的表达有关。
Objective To observe the effect of sour puer ping stomach (SPPW) on invasion and metastasis of human gastric cancer cell line SGC-7901. Methods After SPPW-containing serum was given to human gastric cancer cell line SGC-7901 for 48h, the effect of SPPW on invasion and metastasis of human gastric cancer cell SGC-7901 was tested by adhesion test, invasion assay and migration assay. The effects of SPPW on the expression of MMP-2 and MMP-9 mRNAs related to invasion and metastasis were detected by real-time PCR. Results After intervention of SPPW for 48 h, the adhesion inhibition rate, invasion inhibition rate and migration inhibition rate of SGC-7901 cells increased and in a dose-dependent manner, and there was a significant difference compared with the negative control group (P < 0.01 or P <0.05). There was no significant difference in the expression of MMP-2mRNA among the three dose groups of SPPW. The expression of MMP-9mRNA was statistically different (P <0.01). Conclusion SPPW can inhibit the adhesion, invasion and migration of human gastric cancer cell SGC-7901 in a dose-dependent manner. The mechanism may be related to the down-regulation of MMP-9 expression.
