目的:观察消旋、左旋、右旋丁基苯酞(dl,l,d-NBP)对体外培养的脑血管内皮细胞(BCEC)及主动脉内皮细胞(BAEC)合成一氧化氮(NO),前列环素(epoprostenol,Epo),内皮素-1(ET-1)的影响。方法:自牛大脑灰质及胸主动脉分离培养内皮细胞,并用分光光度法和放免法测定细胞培养液中NO,Epo,ET-1的含量。结果:BCEC及BAEC可持续分泌NO,Epo,ET-1,dl,l-NBP可以剂量依赖性地明显促进BAEC及BCEC中NO的合成,不同浓度的d-NBP均能增加BAEC中NO的产生,而对BCEC无明显作用。低浓度的dl,l-NBP(0.1-10μmol·L~(-1))即可显著升高BCEC中Epo的产生,而对BAEC无明显影响。另外,dl,l,d-NBP对TNFα诱导的BAEC中ET-1增加均无显著影响。结论:1)dl-和l-NBP均可显著增加BAEC及BCEC中NO的释放。2)l-NBP可升高BCEC和BAEC中Epo的释放,且对BCEC具有选择性。dl-NBP选择性增加BCEC中Epo的释放。
OBJECTIVE: To observe the effects of racemic, levo and d-NBP on the synthesis of nitric oxide (NO) in cultured human vascular endothelial cells (BCEC) and aortic endothelial cells (BAEC) Epoprostenol (Epo), endothelin-1 (ET-1). Methods: Endothelial cells were isolated and cultured from bovine cerebral gray matter and thoracic aorta. The contents of NO, Epo and ET-1 in cell culture medium were determined by spectrophotometry and radioimmunoassay. Results: The sustained secretion of NO, Epo, ET-1, dl and l-NBP by BCEC and BAEC significantly promoted the NO synthesis in BAEC and BCEC in a dose-dependent manner. D-NBP at different concentrations increased NO production in BAEC , While no significant effect on BCEC. Low concentration of dl, l-NBP (0.1-10μmol·L -1) could significantly increase the production of Epo in BCEC, but had no significant effect on BAEC. In addition, dl, l, d-NBP had no significant effect on the increase of ET-1 induced by TNFα in BAEC. Conclusions: 1) Both dl- and l-NBP can significantly increase NO release in BAEC and BCEC. 2) l-NBP increases the release of Epo in BCECs and BAECs and is selective for BCECs. dl-NBP selectively increases the release of Epo in BCECs.
