目的:探讨WWOX(WW domain containing oxidoreductase)基因的表达与大肠癌生物学行为及Bax(Bcl-2-associated X protein)、E2F1(E2F transcription factor 1)表达的相关性。方法:应用免疫组化Envision二步法检测58例大肠癌中WWOX、Bax及E2F1的表达,并以20例大肠粘膜炎症为对照。结果:(1)WWOX、Bax在大肠癌中的阳性率分别为25.86%、58.62%,显著低于大肠粘膜炎症的65%、85%,差异有统计学意义。(P<0.01,P<0.05);E2F1在癌组织中的阳性率为63.79%,显著高于炎症组的35%,差异有统计学意义(P<0.05)。(2)E2F1在大肠癌中的表达与浸润深度有关(P<0.05);WWOX、Bax在大肠癌中的表达与患者的预后相关,差异有统计学意义(P<0.01,P<0.05),且WWOX的表达在癌组织的浸润深度间差异有统计学意义(P<0.05)。(3)WWOX与E2F1的表达无相关性(χ~2=2.301,P>0.05),而与Bax的表达有关(χ~2=6.561,P<0.05)。结论:WWOX、Bax及E2F1参与调控大肠癌的发生、发展,且WWOX与Bax协同作用,在大肠癌的预后中扮演了重要角色。
OBJECTIVE: To investigate the correlation between the expression of WWOX-containing oxidoreductase (WWOX) gene and the biological behavior of colorectal cancer and the expressions of Bcl-2 and E2F1. Methods: The expressions of WWOX, Bax and E2F1 in 58 cases of colorectal cancer were detected by immunohistochemical Envision two-step method and 20 cases of colorectal mucosal inflammation as control. Results: (1) The positive rates of WWOX and Bax in colorectal cancer were 25.86% and 58.62%, respectively, which were significantly lower than 65% and 85% of colorectal mucosal inflammation. The difference was statistically significant. (P <0.01, P <0.05). The positive rate of E2F1 in cancer tissue was 63.79%, which was significantly higher than that in inflammatory group (35%), the difference was statistically significant (P <0.05). (2) The expression of E2F1 in colorectal cancer was related to the depth of invasion (P <0.05). The expression of WWOX and Bax in colorectal cancer was correlated with the prognosis of patients (P <0.01, P <0.05) The WWOX expression in the depth of invasion of cancer tissue was significantly different (P <0.05). (3) There was no correlation between WWOX and E2F1 expression (χ ~ 2 = 2.301, P> 0.05), but correlated with the expression of Bax (χ ~ 2 = 6.561, P <0.05). CONCLUSION: WWOX, Bax and E2F1 are involved in the genesis and progression of colorectal cancer. WWOX and Bax synergistically play an important role in the prognosis of colorectal cancer.
