目的:探讨子痫前期(PE)大鼠胎盘组织白细胞介素-6(IL-6)表达异常对胎鼠肺发育的影响。方法:将16只孕鼠随机分为模型组(PE早产组)和对照组(非PE早产组)各8只,模型组使用内毒素建立PE动物模型,对照组给予等量生理盐水。两组孕鼠妊娠第20天时剖腹产取出胎鼠及胎盘,采用免疫组织化学SABC染色法和免疫印迹法检测胎盘组织IL-6表达水平。两组胎鼠饲养14 d后取肺组织制成切片,进行苏木精-伊红(HE)染色,放射状肺泡计数(RAC)分析肺泡发育情况,免疫印迹法检测肺组织血管内皮生长因子(VEGF)表达水平。结果:模型组孕鼠胎盘组织IL-6表达水平高于对照组,胎鼠RAC多于对照组,胎鼠肺组织VEGF表达水平高于对照组(P均<0.05)。结论:PE大鼠胎盘组织IL-6的表达增高,进而促进胎鼠肺组织VEGF的表达和肺泡的形成,但胎鼠肺泡缺乏表面活性蛋白而导致肺发育不成熟。
Objective: To investigate the effect of abnormal expression of interleukin-6 (IL-6) on the development of fetal rat lung in preeclampsia (PE) rats. Methods: Twenty-six pregnant rats were randomly divided into model group (PE preterm group) and control group (non-PE premature group), each group had 8 rats. Endotoxin was used to establish animal model of PE in model group and normal saline in control group. The fetus and placenta were removed by cesarean section on the 20th day of gestation in both pregnant rats. The expression of IL-6 in the placenta was detected by immunohistochemical SABC staining and immunoblotting. Two groups of fetal rats were kept for 14 days and then the lungs were obtained and stained with hematoxylin and eosin (HE). Radial alveolar count (RAC) was used to analyze the development of alveoli. Western blotting was used to detect the expression of vascular endothelial growth factor )The expression level. Results: The expression level of IL-6 in placental tissue of model group was higher than that of control group. The expression level of VEGF in fetal rat lung was higher than that in control group (all P <0.05). CONCLUSION: The expression of IL-6 in placental tissue of PE rats increases, which in turn promotes the expression of VEGF and the formation of alveoli in the fetal rat lungs. However, the absence of surfactant protein in fetal rat alveoli leads to immature lung development.
