目的:探讨丹红注射液对大鼠心肌缺血再灌注(I/R)后血清炎症因子的影响。方法:30只雄性SD大鼠随机分为假手术组(10只)、模型组(10只)、药物组(10只)。模型组和药物组大鼠通过穿线结扎左冠状动脉前降支与松开的方法制备心肌缺血再灌注损伤模型(心肌缺血30min,再灌注2h)。假手术组只穿线不结扎。药物组大鼠在模型建立前5天开始每天给予丹红注射液2mL/kg腹腔注射,假手术组和模型组只给予同等剂量的生理盐水腹腔注射。各组心肌缺血30min,再灌注2h采血离心取上清采用ELASA的方法检测血清中ICAM-1及白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、的水平。结果:与假手术组比较,模型组大鼠的IL-1β、IL-6、TNF-α、ICAM-1水平明显升高(P均<0.05);与模型组相比,药物组大鼠的IL-1β、IL-6、TNF-α、ICAM-1水平明显降低(P均<0.05)。结论:丹红注射液可以降低心肌缺血再灌注损伤的黏附分子1(ICAM-1)及炎症因子水平,具有心肌保护作用。
Objective: To investigate the effect of Danhong injection on serum inflammatory factors after myocardial ischemia reperfusion (I / R) in rats. Methods: Thirty male SD rats were randomly divided into sham operation group (n = 10), model group (n = 10) and drug group (n = 10). The model of myocardial ischemia and reperfusion injury was established by ligating the left anterior descending coronary artery and loosening the rats in model group and drug group (myocardial ischemia 30min, reperfusion 2h). Sham operation group only threading not ligation. The rats in the drug group were given intraperitoneal injection of Danhong injection 2mL / kg every day 5 days before model establishment. The sham operation group and model group were given intraperitoneal injection of the same dose of saline only. Each group of myocardial ischemia 30min, 2h reperfusion after centrifugation to take the supernatant by ELASA method for detection of serum ICAM-1 and interleukin -6 (IL-6), interleukin-1β (IL-1β), tumor necrosis Factor-α (TNF-α) levels. Results: Compared with sham operation group, the levels of IL-1β, IL-6, TNF-α and ICAM-1 in model group were significantly increased (all P <0.05). Compared with model group, The levels of IL-1β, IL-6, TNF-α and ICAM-1 were significantly decreased (all P <0.05). Conclusion: Danhong injection can reduce myocardial ischemia-reperfusion injury adhesion molecule 1 (ICAM-1) and inflammatory cytokines levels, with myocardial protection.
