目的 探讨牛磺酸和复合微量营养素对光照和暗适应条件下大鼠视网膜代谢型谷氨酸受体 6亚型 (m Glu R6) m RNA的影响。方法 在幼年大鼠饲料中添加牛磺酸或 VA、VB1、VB2 、尼克酸、Zn、Se等复合微量营养素 ,营养干预 4wk后 ,采用原位分子杂交技术检测各组大鼠视网膜 m Glu R6m RNA在光照和暗适应条件下的分布和表达。结果 光照和暗适应时 m Glu R6m RNA均集中表达于内、外核层和神经节细胞层。牛碘酸能增加光照和暗适应时上述部位m Glu R6m RNA的表达 ,而复合微量营养素对其表达无明显影响。结论 除双极细胞外 ,部分神经节细胞也表达 m Glu R6;光感受器突触前膜存在反馈性的 m Glu R6。牛磺酸可能通过促进光照和暗适应条件下 m Glu R6m RNA的表达 ,调制大鼠视杆通路视觉信号传递
Objective To investigate the effects of taurine and micronutrients on m Glu R6 mRNA in rat retina under light and dark adaptation. Methods The micronutrients such as taurine, VA, VB1, VB2, niacin, Zn and Se were added into juvenile rats’ diet. After nutritional intervention for 4 weeks, the in situ hybridization was used to detect the expression of m Glu R6mRNA Distribution and expression under light and dark adaptation conditions. Results The m GluR6m RNAs were concentrated in the inner and outer nuclear layers and the ganglion cell layers under both light and dark adaptation. Cattle iodine increased the expression of mGluR6mRNA in the above sites when exposed to light and dark, while the composite micronutrients had no significant effect on its expression. Conclusion In addition to bipolar cells, some ganglion cells also express m Glu R6; there is feedback m Glu R6 in the presynaptic membrane of photoreceptors. Taurine may modulate the visual signal transmission in rat’s rod pathway by promoting the expression of mGluR6mRNA under both light and dark adaptation
