目的探讨急、慢性白血病治疗前骨髓塑料包埋切片内cmyc原癌基因表达的变化。方法使用冷丙酮固定结合简易三步塑包体系,观察了22例急、慢性白血病患者骨髓活检GMA包埋切片内白血病细胞cmyc表达的检测结果。结果急性髓系白血病(AML)时,19%~69%白血病细胞内含有cmyc蛋白,其含量在每例患者中多变。比较之下,治前的急性淋巴细胞白血病(ALL)和慢性髓系白血病(CML)慢性期中就罕有检出cmyc阳性细胞者。但2例CML加速期患者标本白血病细胞内cmyc蛋白表达增高。结论本研究建立的一种骨髓活检标本的简易塑包操作技术,既可获卓越的抗原性保留,也能高质量地对白血病细胞内cmyc蛋白表达的形态学细节进行观察。
Objective To investigate the changes of cmyc proto-oncogene expression in plastic bone marrow-embedded sections before and after acute and chronic leukemia treatment. Methods The results of c-myc expression in leukemia cells in bone marrow biopsy GMA-embedded sections of 22 acute and chronic leukemia patients were observed using cold acetone fixation combined with a simple three-step plastic bag system. Results In acute myeloid leukemia (AML), 19% to 69% of the leukemia cells contained c-myc protein, and the content was variable in each patient. In comparison, precancerous patients with acute lymphoblastic leukemia (ALL) and chronic myeloid leukemia (CML) rarely detect c-myc-positive cells during the chronic phase. However, the expression of cmyc protein in leukemia cells was increased in 2 patients with CML in accelerated phase. Conclusion The simple plastic packaging technique of bone marrow biopsy specimens established in this study can not only obtain excellent antigenic retention, but also can observe the morphological details of cmyc protein expression in leukemia cells with high quality.
