目的 研究丙型肝炎病毒(HCV)核酸疫苗NV-HC/NS3接种BALB/c 小鼠后,对相应靶抗原攻击的特异性免疫预防和治疗效应、方法以HCV核酸疫苗NV-HC/NS3接种BALB/c小鼠,分别在疫苗接种前后种植能表达靶抗原的BALB/c 小鼠骨髓瘤细胞株SP2/0,观察疫苗接种对肿瘤生长及小鼠寿命的影响。结果种植肿瘤前先接种NV-HC/NS3的小鼠,荷瘤阻抑率达40%;长出肿瘤的小鼠与未接种疫苗的对照组小鼠相比,生存时间延长,生存率提高。先荷瘤后免疫的小鼠,核酸疫苗的接种则能提高小鼠的生存率。结论NV-HC/NS3能诱导小鼠产生针对靶抗原的特异性免疫应答,发挥预防甚至治疗效应。
Objective To study the specific immunological prophylaxis and therapeutic effect of vaccination against hepatitis B virus (HCV) vaccine NV-HC / NS3 on BALB / c mice. Methods BALB / c mice were vaccinated with HCV DNA vaccine NV-HC / NS3 / C mice were immunized with BALB / c mouse myeloma cell line SP2 / 0 before and after vaccination to observe the effect of vaccination on tumor growth and life span of mice. Results Inoculation of NV-HC / NS3 mice before tumor implantation resulted in a 40% tumor-inhibition rate. Compared with non-vaccinated control mice, tumor-bearing mice showed prolonged survival and improved survival rate. Mice that were immunized with the first tumor and vaccinated with the nucleic acid vaccine could improve the survival rate of the mice. Conclusion NV-HC / NS3 can induce mice to produce specific immune response against target antigen and exert preventive and even therapeutic effects.
