目的 :对申克孢子丝菌进行基因分型研究 ,探索申克孢子丝菌的基因分型标准。方法 :用光镜对 2株标准株 ,4株环境分离株和 2 4株临床分离菌株进行形态特征观察 ,再用随机扩增DNA指纹方法 (RAPD)对DNA进行PCR扩增 ,根据扩增产物的电泳带型来分析DNA多态性。结果 :30株菌株镜下可见 2种形态结构 ;30株申克孢子丝菌的DNA指纹带型不同 ;多引物聚类分析表明 ,3条引物可将 30株不同地区来源申克孢子丝菌分作 11个型。结论 :(1)在我国申克孢子丝菌中存在 2种不同的形态结构。 (2 )申克孢子丝菌中存在不同的基因型。 (3)RAPD法用于申克孢子丝菌菌株鉴定是一种快速、方便、可行的方法。
OBJECTIVE: To study the genotyping of Spirorhizospora schrenckii and explore the genotyping criteria of Sporothrix schenckii. Methods: The morphological characteristics of 2 standard strains, 4 environmental isolates and 24 clinical isolates were observed by light microscopy. The DNA was amplified by random amplified DNA fingerprinting (RAPD). According to the amplification products Of the electrophoresis band to analyze DNA polymorphisms. Results: There were two kinds of morphological structures in 30 strains of microspheres. The genotypes of DNA fingerprints in 30 strains of Spirorubaci were different. The multi-primer clustering analysis showed that three primers could separate 30 strains of sporotrichum from different regions For 11 types. Conclusions: (1) There are two kinds of morphological structures in Sporotrichosis in China. (2) There are different genotypes in Spirulina. (3) RAPD method for the identification of Spirulina conidia strains is a fast, convenient and feasible method.
