目的:观察益气养阴方对早期糖尿病肾病大鼠的疗效并探讨其作用机制。方法:48只Wistar大鼠随机分为正常组、模型组、益气养阴方组、西药组。除正常组外,其余组均采用单侧肾脏切除、高脂高糖饮食联合STZ制备糖尿病肾病大鼠模型,观察各组大鼠治疗后血糖、尿白蛋白、血肌酐、尿素氮、单核细胞趋化蛋白-1(MCP-1)mRNA的变化。结果:模型组大鼠血糖、尿白蛋白较正常组高(P<0.05),益气养阴方组和西药组大鼠各项指标均好转,其中益气养阴方组血糖值及西药组血糖值、尿白蛋白值与模型组比较,差异有统计学意义(P<0.05);模型组肾脏组织内MCP-1 mRNA的表达较正常对照组高(P<0.05),益气养阴方干预后其表达下降(P<0.05)。结论:益气养阴方能减少糖尿病肾病大鼠血糖及尿白蛋白值,该作用可能与下调MCP-1 mRNA的表达有关。
Objective: To observe the effect of Yiqi Yangyin Recipe on early diabetic nephropathy rats and to explore its mechanism. Methods: Forty-eight Wistar rats were randomly divided into normal group, model group, Yiqi Yangyin prescription group and western medicine group. In addition to the normal group, the other groups were treated with unilateral nephrectomy, high-fat and high-sugar diet combined with STZ diabetic rat model of diabetic nephropathy, blood glucose, urinary albumin, serum creatinine, urea nitrogen, monocytes Changes of chemotactic protein-1 (MCP-1) mRNA. Results: The levels of blood glucose and urine albumin in model group were significantly higher than those in normal group (P <0.05). The indexes of Yiqi Yangyin prescription group and western medicine group were all improved. The blood sugar level of Yiqi Yangyin prescription group and western medicine group (P <0.05). The expression of MCP-1 mRNA in the kidney of the model group was higher than that of the normal control group (P <0.05). The level of blood glucose and urinary albumin in the model group was significantly higher than that in the model group After intervention, its expression decreased (P <0.05). Conclusion: Yiqi Yangyin Prescription can reduce blood glucose and urine albumin in diabetic nephropathy rats, which may be related to down-regulating the expression of MCP-1 mRNA.
