目的探究miR-93的表达与其宿主基因MCM7的表达的相关性。方法脂肪组织来源于16例患多囊卵巢综合征(PCOS)的妇女,其中8例有胰岛素抵抗(IR),8例无IR。另选15例非PCOS患者(9例有IR,6例无IR)。检测脂肪组织细胞中MCM7和miR-93的表达。结果 MCM7在非PCOS/IR和PCOS女性中的表达量降低,并且往往在PCOS和IR女性患者中表达量降低(P<0.05)。人类脂肪组织中MCM7的表达与miR-93的表达呈负相关,而与受试者IR的增加呈正相关(P<0.05)。脂肪细胞外胰岛素的增加对MCM7或miR-93的表达均无影响(P>0.05)。结论MCM7和miR-93的表达与PCOS和IR有关。虽然葡萄糖水平升高能下调MCM7的表达,但是脂肪组织中高胰岛素血症或高糖血症均不能改变miR-93表达。miR-93表达调控机制可能与其宿主基因MCM7的机制有明显区别。
Objective To investigate the relationship between the expression of miR-93 and the expression of its host gene MCM7. Methods Adipose tissue originated from 16 women with polycystic ovary syndrome (PCOS), of whom 8 had insulin resistance (IR) and 8 had no IR. Another 15 non-PCOS patients were selected (9 with IR and 6 without IR). The expression of MCM7 and miR-93 was detected in adipose tissue cells. Results The expression of MCM7 decreased in non-PCOS / IR and PCOS women, and often decreased in PCOS and IR female patients (P <0.05). The expression of MCM7 in human adipose tissue was negatively correlated with the expression of miR-93, but positively correlated with the IR of the subjects (P <0.05). Increased extracellular fat insulin had no effect on the expression of MCM7 or miR-93 (P> 0.05). Conclusion The expressions of MCM7 and miR-93 are correlated with PCOS and IR. Although elevated glucose levels down-regulated the expression of MCM7, neither hyperinsulinemia nor hyperglycemia in adipose tissue altered miR-93 expression. The mechanism of miR-93 expression may be significantly different from the mechanism of its host gene MCM7.
