目的 :本实验采用高效液相色谱法 ,以离体小白鼠腹部皮肤为透皮屏障 ,比较了四种双氯灭痛软膏剂的透皮效果。方法 :实验用法国Gilson公司的HPLC仪 ,以C18为固定相 ,甲醇 水 (含HAc 0 .36 % ) =80∶2 0为流动相 ,紫外检测波长 2 80nm。平均回收率为 99.5 9% ,RSD <3.2 8% ,检测限为 1.0 μg/ml。 结果 :1号样品透皮率最高 ,3号与 1号相比较透皮吸收无显著性差异(P >0 .0 5 ) ,2号或 4号分别与 1号相比 ,其透皮吸收均有显著性差异 (P <0 .0 5 )。结论 :1号样品值得进一步开发。
OBJECTIVE: To study the transdermal effect of four diclofenac ointments in this experiment by using high performance liquid chromatography (HPLC) with the abdominal skin of isolated mice as the transdermal barrier. Methods: The experiment was performed with a Gilson HPLC instrument using C18 as the stationary phase and methanol water (containing HAc 0.36%) = 80: 20 as the mobile phase. The UV detection wavelength was 280 nm. The average recovery was 99.5 9%, RSD <3.2 8%, and the detection limit was 1.0 μg / ml. Results: The transdermal absorption of No.1 sample was the highest, while there was no significant difference in transdermal absorption between No.3 and No.1 (P> 0.05). The transdermal absorption of No.2 or No.4 was There was a significant difference (P <0.05). Conclusion: Sample No. 1 deserves further development.
