目的应用SHSY5Y神经母细胞瘤建立脑干肿瘤裸鼠模型。方法将SHSY5Y神经母细胞瘤细胞注射到裸鼠脑桥,2.5周后行MRI扫描观察肿瘤生长情况,行苏木精-伊红染色、免疫组化染色观察肿瘤细胞形态及胶质纤维酸性蛋白(GFAP)、巢蛋白(Nestin)和神经蛋白丝(NF)的表达。结果 MRI检查显示脑干部位出现肿瘤,为长Tl、长T2信号,注射钆喷酸葡胺(Gd-DTPA)后信号明显增强。苏木精-伊红染色显示肿瘤为神经母细胞瘤,呈浸润性生长,细胞围绕毛细血管排列成假菊形团结构。免疫组化显示GFAP及NF呈阴性表达,Nestin呈阳性表达。结论 SHSY5Y神经母细胞瘤可用于建立脑干肿瘤动物模型,该模型具有稳定的重复性和可预测性。
Objective To establish a brain stem tumor model in nude mice using SHSY5Y neuroblastoma. Methods SHSY5Y neuroblastoma cells were injected into the pons of the nude mice. The growth of the tumors was observed by MRI scanning after 2.5 weeks. The hematoxylin-eosin staining and immunohistochemistry staining were used to observe the morphology of tumor cells and glial fibrillary acidic protein (GFAP) Nestin and Neurofilament (NF) expression. Results MRI examination showed that there was tumor in the brain stem area, which was a long Tl and long T2 signal. The signal of Gd-DTPA injection was significantly enhanced. Hematoxylin-eosin staining showed that the tumor was neuroblastoma, which showed invasive growth, and the cells arranged in a pseudo-daisy-like structure around the capillaries. Immunohistochemistry showed negative expression of GFAP and NF, Nestin positive expression. Conclusion SHSY5Y neuroblastoma can be used to establish animal model of brain stem tumor. The model has stable repeatability and predictability.
