目的研究Notch1、P21WAF1/Cip1及活化caspase3在肝细胞癌中表达的相关性。方法利用免疫组织化学检测60例肝细胞癌(HCC)和15例正常肝组织中Notch1、P21WAF1/Cip1及活化caspase3的表达。结果HCC中Notch1、P21WAF1/Cip1及活化caspase3的阳性率和阳性表达强度显著低于正常肝组织(P<0.01);Notch1、P21WAF1/Cip1及活化caspase3的表达强度与HCC分级和肿瘤大小密切相关,肿瘤分化愈差和肿瘤直径愈大,3种蛋白表达愈弱(P<0.01);在HCC中,P21WAF1/Cip1和活化caspase3的表达强度与Notch1表达强度呈显著正相关(r分别为0.76和0.71)。结论提示Notch1信号通路可能是通过影响细胞周期和细胞凋亡在HCC发生发展过程中发挥负性调控作用。
Objective To investigate the expression of Notch1, P21WAF1 / Cip1 and activated caspase3 in hepatocellular carcinoma. Methods Immunohistochemistry was used to detect the expressions of Notch1, P21WAF1 / Cip1 and activated caspase3 in 60 cases of hepatocellular carcinoma (HCC) and 15 cases of normal liver tissue. Results The positive rate and positive expression of Notch1, P21WAF1 / Cip1 and activated caspase3 in HCC were significantly lower than those in normal liver tissue (P <0.01). The expression of Notch1, P21WAF1 / Cip1 and activated caspase3 were closely related to the grade of HCC and the size of tumor, The poorer the tumor differentiation and the larger the tumor diameter were, the weaker the expression of three proteins (P <0.01). In HCC, the expressions of P21WAF1 / Cip1 and activated caspase3 were positively correlated with the expression of Notch1 (r = 0.76 and 0.71 ). Conclusions suggest that Notch1 signaling pathway may play a negative regulatory role in the development and progression of HCC by affecting cell cycle and apoptosis.
